The known side effects of IGF-1 injections include jaw pain, facial and hand swelling and heart-rhythm disturbances, especially if doses of more than 100 micrograms (mcg) are injected. Exceeding 100mcg of IGF-1 can actually cause your heart to stop beating and blood pressure to drop dramatically. This is caused by an IGF-1-induced drop in blood phosphate levels, and in the bodybuilding community is often prevented by administering phosphate with the IGF-1.

It is extremely important to create awareness among the masses, that procuring illegal drugs off the black market or the internet is an offence under the federal law, being guilty of which can lead to a long jail sentence. Hexarelin via CD36 occupation increases the expression of multiple genes involved in fatty acid mobilization in adipocytes toward the mitochondrial oxidative phosphorylation, and many of these upregulated genes are known targets of PPARγ. Consistent with this, electron microscopy of hexarelin-treated adipocytes reflects highly organized cristae formation that spans the entire width of mitochondria, with a concomitant cytochrome c oxidase activity enhancement. Although this signaling and activation cascade has not been described for myocardial cells so far, the potential existence of these phosphorylative and mitochondriogenic mechanisms in the heart, and its potential amplification by GHRP ligands, may eventually contribute to myocardial salvage during critical ischemia periods.47 In a more recent study based on a myocardial infarction model, and addressed to examine whether hexarelin treatment can compensate for ghrelin deficiency in ghrelin-knockout mice, the mortality within two weeks was significantly lower in the hexarelin (6.7%) and ghrelin groups (14.3%) than in the vehicle group (50%). Furthermore, hexarelin was more effective than ghrelin as judged by the ejection fraction and other LV-dependent physiological constants as dP/dt max and dP/dt min, which is a measure of LV global contractility.48


GHRP-6 is a peptide from the category of growth hormone releasing peptides, ie growth hormone-releasing peptides. The most frequent use of these peptides is an extreme increase in the body's growth hormone production. The main application area of GHRP-6 is an increase in growth hormone levels, which also results in an increase in IGF1 levels. This also radically supports fat loss as well as muscle building. Cancer can often be a process of uncontrolled cellular division. IGF-1 is not only pro-growth in a way that could increase this cellular division, but IGF-1 also inhibits apoptosis, or programmed cell death. Hence the theory among some in the medical community that tumors could increase synthesis of IGF-1 to keep themselves alive and to encourage the spread of cancer throughout the body. This doesn’t mean that IGF-1 directly causes cancer.
Remember the GHRP you select is used for a few reasons. One is to prompt the release of the increase pulse in GH you have initiated with the GHRH you have selected to use. This is by inhibition of Somatostatin. So you are actually selecting the timing of the release of your natural production of  still physiologic amount of GH.  Another reason is to actually contribute a little more to the amplitude of you GH pulse.

CJC 1295 can be compounded in two forms (DAC and non-DAC). Drug affinity complex (DAC) prevents enzymatic degradation thus increasing the half-life. Consequently CJC 1295-DAC can be dosed as a single weekly injection. Administration of CJC 1295-DAC provides a GHRH-like stimulation around the clock. A potential drawback when using a weekly protocol can be attributed to ineffective GHRH stimulation when the body is due for a GH spike (usually 1:00am). This is referred to as a GH-bleed and the overall result is inferior to using CJC 1295-NON-DAC daily for 5 days out of 7. Therefore using CJC 1295-NON-DAC daily (between 6-8pm) provides a more effective GH spike at 1:00am.
Furthermore, the most potent profibrogenic growth factors: Tgfb1, Pdgfb, and Ctgf also appeared significantly underexpressed in the GHRP-6-treated wounds (all ) (Figure 4). In line with this, we observed a significant reduction in the expression levels of Col1a1 and Col3a1 (Figure 4, both ). Concomitantly, we addressed the attention to filamentous and contractile proteins associated with fibroblasts and other differentiated mesenchyme-derived cells. Acta2 appeared close to a significant reduction (), whereas Des, Vim, and Fn transcriptional expression appeared significantly reduced (all ), as compared to placebo-treated wounds.
One of the major differences between GHRP 2 and GHRP 6 is that the latter increases hunger in you substantially, especially when you consume the supplement at regular intervals. Therefore, those looking to build muscles and lose excess fat may want to consider GHRP 2 as it is not known to build appetite in you to that extent. However, if your aim is to eat more and growth quickly then GHRP 6 based supplements is for you.
Ghrelin is a potent stimulator of growth hormone secretion from the anterior pituitary gland. The ghrelin receptor is a G protein-coupled receptor, known as the growth hormone secretagogue receptor. Ghrelin binds to the GHSR1a splice-variant of this receptor which is present in high density in the hypothalamus, pituitary as well as vagal afferent cell bodies and vagal afferent endings throughout the gastro-intestinal tract.

Statistical analyses were carried out using GraphPad Prism 6 for Windows, version 6.01. For clinical response, histomorphometric parameters, and gene expression data, normal distribution (Kolmogorov-Smirnov) and variance homogeneity (Brown-Forsythe) tests were performed. Once normality was demonstrated, differences between GHRP-6-treated and placebo-treated animals were determined using two-tailed unpaired Student’s -test. For non-Gaussian distributed data, Mann-Whitney U test was performed. For analyzing closure kinetics of rat wounds, two-way ANOVA was performed, followed by Sidak’s multiple comparisons test. In all cases, values of were considered statistically significant. The values shown represent mean ± SD (error bars).
As previously explained, multiple GHRP-6 doses are required throughout the day due to the pulsatile nature of the HGH release, and the administration of these doses are typically administered on average 3 times daily spaced evenly apart. More administrations are acceptable for greater effects on physique and performance, but it is advised that approximately 3 hours in between each injection is ensured so as to allow the pituitary gland to restore its storage of HGH. Well, personally I used 150mcg injected directly into the joints or areas that I've had any niggling injuries, the localized effect it has on collagen growth is nothing short of astounding. After using GHRP-6, I have personally recovered from a full pectoral tendon tear, where the tendon ripped right of the humerus bone; in fact, it's now in better shape than it was prior to the injury. Interestingly, 5g of the GHRP-6 will last ages when used properly; even at this dose fat loss is noticeable and the anabolic effects of increased muscle size and strength can be seen.
A remarkable specific (125)I-Tyr-Ala-hexarelin binding was observed in the human cardiovascular system where the highest binding levels were detected in ventricles, followed by atria, aorta, coronaries, carotid, endocardium, and vena cava. In other experiments on H9c2, cardiomyocyte-specific GHRP binding was found along with a potent antiapoptotic activity.3 The primarily investigated receptor was the growth hormone secretagogue receptor type 1a (GHS-R1a), which was detected in isolated human cardiomyocytes, myocardium, and aorta samples.17 It has been recently shown that GHS-R1a is a sort of “promiscuous receptor” involved in many systems and behavioral patterns such as reward, feeding, and memory, which makes it an attractive pharmacological target.18 Years later, the synthetic GHRP hexarelin was acknowledged as a ligand of another protein identified as CD36, a scavenger receptor that is expressed in various tissues, including monocytes/macrophages and the endothelial microvasculature. Activation of CD36 in perfused hearts by hexarelin was shown to increase coronary perfusion pressure in a dose-dependent manner. Contrariwise, this effect was lacking in hearts from CD36-null mice and hearts from spontaneous hypertensive rats genetically deficient in CD36.19,20 Thus, it is currently accepted that two cardiac receptor subtypes mediate the pharmacological actions of GHRP-6, GHRP-2, and hexarelin.21,22
From the standpoint of protein synthesis and muscle repair, IGF-1 injections have also been shown to enhance the anticatabolic effects of insulin and to increase the protein synthesis normally induced by growth hormone. This is because, like insulin, IGF-1 encourages amino acid uptake into muscle cells, stimulates peripheral tissue uptake of glucose (which lowers blood glucose levels), and suppresses liver glucose production. That last fact is important and is actually why IGF-1 is even being considered as a diabetes-prevention drug. Insulin resistance can cause the liver to produce excess glucose, which then causes even more insulin insensitivity and can eventually result in type II diabetes, and IGF-1 can decrease the need for this type excessive insulin release.
Of particular note is the variable chemistry of GHRPs, which consist of three major chemical classes including peptides, partial peptides, and nonpeptides, all of which appear to act via the same receptor and cellular mechanisms. Generally, most GHRPs are active by all routes of administration, specifically intravenously (IV), subcutaneously (SC), orally, intranasally, and intracerebroventricularly (IVC), which supports their possible broad future clinical utility. From evolutionary studies starting with the zebrafish, the natural receptor and hormone have been present for hundreds of years, underscoring the fundamental evolutionary and functional importance of the ghrelin system. GHRPs were well established to act directly on both the hypothalamus and pituitary several years before the GHS receptor assay.23
Results in Fig. 1.8 of normal young men (left panel) and women (right panel) demonstrate that iv bolus combined GHRP-2 and GHRH at the respective doses of 1 μg/kg GHRH and a subthreshold GH-releasing dose of 0.03 μg/kg GHRP-2 released GH synergistically (Bowers, 1998). From these studies, GHRP is envisioned to act on the hypothalamus to release an unknown factor (U factor) rather than endogenous GHRH which subsequently acts concomitantly with GHRH on the pituitary somatotroph to release GH synergistically. In this study, the important specific finding is that GHRP-2 augments GHRH release even when GHRH is present in excess amounts, and the concomitant GHRP-2 dose of 0.03 μg/kg is a subthreshold GH-releasing amount. Thus, GHRP + GHRH is not releasing GH in this study by augmenting endogenous GHRH release and, furthermore, GHRP+GHRH release in vitro is additive and not synergistic. In addition, from other high-dosage GHRP-2 data, that is, 10 μg/kg sc (not shown), we have postulated that at high doses GHRPs do act on the hypothalamus to release endogenous GHRH because high-dose GHRP-2 (10 μg/kg sc) releases the same large amount of GH released by combined GHRH + GHRP-2 at 1 + 1 μg/kg iv (Bowers, 1998a,b).
Finally, the ghrelin chemical isolation and identification was accomplished surprisingly from the stomach, which is the major site but not the only site. Ghrelin was isolated and identified.4 A primary action of GHRPs continues to concern GH secretion and regulation, but increasingly this has included direct and indirect effects on nutrition and metabolism, as well as a variety of other actions which may be pharmacological and/or physiological.
Bremelanotide PT 141 was developed from Melanotan II, targeting its aphrodisiac effects. This peptide has been shown to have a substantial effect on libido, generating sexual arousal in both men and women within minutes of administration. It has been shown to be effective in treating erectile dysfunction, even in men who have not responded to other ED treatments, such as Viagara. This peptide is also able to cross the blood-brain-barrier, bypassing the vascular system and acting at the level of the central nervous system. This property gives Bremelanotide an advantage over traditional ED drugs, which can decrease blood pressure to dangerous levels. This peptide can be administered as a nasal spray, making its use convenient and discreet.

Everybody has unique goals and these are best adjusted by your dosages. Research in The Journal Of Clinical Endocrinology & Metabolism found 100mcg will saturate all your receptors, but taking 200mcg will cough up an additional 50% of effectiveness, where as 300mcg delivers just a 25% of an additional boost. So the law of diminishing returns is firmly in place with this peptide. What’s more, higher doses were found in a study in The Journal Of Clinical Endocrinology & Metabolism to increase people’s stress hormone, cortisol in doses over a 100mcg so if you do decide to delve into this supplement, stick to the lower doses.
