At the histological analysis, and from a qualitative perspective, these wounds appeared less inflamed and with a higher degree of ECM organization, given by far less fibrin accumulation and thinner and horizontally distributed collagen bundles. Vessels were also aligned with the collagen fibers. Thus, the treatment not only reduced the wound area but also appeared to be associated with differences in the quality of the ECM as the inflammatory infiltrate. Figure 2(a) is representative of the GHRP-6 effect on the inflammatory response, illustrating the reduction of infiltrated cells as compared to placebo-treated wounds (Figure 2(b)).
Following the preliminary histological data, suggesting a reduction of wound inflammation and a far more organized ECM, we addressed the gene expression study toward inflammatory and profibrogenic markers. We primarily examined Cd36 expression following topical GHRP-6 application and found that peptide reduced its receptor expression () (Figure 4). Furthermore, the treatment significantly reduced Adam17 expression () and approached to significantly reduce Tnf (), which may partially contribute to explaining the substantial reduction of infiltrated inflammatory cells within the wound bed (Figure 4).
The peptide therapy protocols (Amino Acid Analogs) prescribed by TeleWellnessMD providers are also known as secretagogues (pronounced se-creta-gog), a substance that promotes secretion. These amino acid chains communicate with the body to produce or release growth hormone. Hence a secretagogue causes the body’s own natural processes to produce growth hormone. Secretagogues do not act as growth hormones but rather stimulate the pituitary gland to secrete your stored growth hormone. The subcutaneous injection route of growth hormone stimulation is a preferred route to help slow down age and environmental reductions in growth hormone levels.
If you are an athlete or a bodybuilder looking for ways to decrease body fat whilst increasing muscle mass then GHRP 2 and GHRP 6 (the two types of Growth Hormone Releasing Peptides) can certainly help you with your requirements. The best part with these hormones is that you are likely to get the desired results out of them, especially when you combine them with muscle building and fat-burning foods, aerobic and intense strengthening exercises. However, there are subtle differences between the two hormones that you cannot ignore.
Paracetamol/caffeine formulations have a long-established safety and efficacy profile over 25 years of use as an open-sale medicine in major markets around the world. The paracetamol/caffeine combination analgesic was registered as a schedule 2 product in Australia and has been marketed since 2010. Since that time no new significant issues or potential risks have been reported.
The ACMS recommended that Growth Hormone Releasing Hormones (GHRHs), Growth Hormone Secretagogues (GHSs), Growth Hormone Releasing Peptides (GHRPs) as well as new individual substance entries for CJC-1295, ipamorelin, pralmorelin (Growth Hormone Releasing Peptide-2), Growth Hormone Releasing Peptide-6, hexarelin and AOD-9604 be included in Schedule 4.
Melanotan II is an analogue of alpha melanocyte stimulating hormone, the hormone responsible for pigmentation in skin and hair. This peptide has been shown not only to increase skin pigmentation, resulting in a substantially tanner skin tone, but also to stimulate fat loss and increase libido. Its aphrodisiac effects were so substantial that it was the basis for the development of another peptide designed exclusively to address erectile and sexual dysfunction—Bremelanotide PT 141.
When you are just getting started with Ipamorelin, it is advised to use only one supplement daily at the same time each day. It is also advised to begin on the lower end, typically an eight-week cycle, and at a maximum twelve-week cycle. Doing this not only guarantees the desired results when using Ipamorelin, it is also going to ensure you get the most out of the supplement. When using this dosage cycle you will:
Evidence review and acceptance by the NDPSC in 2007, demonstrated that paracetamol/caffeine combination analgesics have a very low risk of nephrotoxicity. Similarly, the combination analgesics pose a very low risk of toxicity in overdosing with only two fatal cases reported in the USA. However, these cases involved other medications in addition to paracetamol/caffeine with the latter being available in very large pack sizes. Further, there are no known contraindications to the paracetamol/caffeine combination apart from hypersensitivity to the constituents."
The final verdict is to go for it. GHRP-6 is not your garden-variety performance enhancer. One of the most important aspects of it is that it does not cause desensitization. This means that it will not cause overdose and that works best for athletes. Also since it increases appetite, it forces you to make more calories available to your body for exercise and for recovery. It does not boost unnatural muscle growth like other performance enhancers and the result does not put extra pressure on your tendons and ligaments.
Steve Coggan, Chatswood, Sydney, “I am 48 and been weightlifting for 20 years. I have used CJC 1295 and Ipamorelin on their own and together, and have to say that when combining them they work better and faster. I have been using them for the past year, with 2 rest times. The combination of CJC 1295 Ipamorelin, is proving fantastic all round. Not only am I increasing muscle, my weight is down, but my skin is looking great. Some of the bags under my eyes have more or less gone. My missus thinks I look 10 years younger! Also, it has helped in the recovery of an injury to my knee after a fall. I have to say it isn’t cheap, but the results are worth it. CJC 1295 Ipamorelin side effects were few, just some headaches at first. You need to use it with a good diet and fitness regime for best results. I will definitely be continuing with it!”
One more way for growth hormone to help with fat loss is that this sustains the levels of blood glucose through inhibiting glucose uptake to the peripheral cells, reducing the glucose oxidation for the energy in cells and thus boosting the production of the glucose in the cells from amino acids and fats. The blood’s free fatty acids from lipolysis also partially obstruct the insulin receptors on the cell membranes, reducing insulin’s effectiveness in triggering glucose removal from the blood that causes decreased sensitivity to insulin or insulin resistance. These will then result to fat loss, particularly from the difficult to move intra-abdominal storages of fat.

Unlike GHRP 6, the Half Life of GHRP 2 is pretty short. In fact, GHRP 2’s peak can be seen occurring within fifteen minutes after it has been administered or at least one hour at the latest. Also unlike GHRP 6, GHRP 2 helps improve the levels of calcium in the body and this can in turn facilitate the secretion of other growth hormones. GHRP 2 is believed to be more potent in its operation as compared to other peptide forms, including GHRP 6. Therefore, it is not surprising to see this peptide commonly employed for treating catabolic deficiencies.
Growth hormone (GH) was first identified for its notable effect on longitudinal growth. Subsequent research revealed that the GH has anabolic effects on protein, lipid, and carbohydrate metabolism. GH replacement therapy, using recombinant GH, is therefore used to treat individuals with short stature due to a variety of conditions. However, GH replacement therapy suffers from significant drawbacks such as low bioavailability and side effects. Moreover, most GH-deficient individuals exhibit a secretory defect rather than a primary deficiency in GH production. Research seeking a better drug to replace GH was therefore extensively active in the 1980s and 1990s.
As the name indicates, this peptide is a fragment of human growth hormone. It is more specifically a modified form of the amino acids 176-191 in the C-terminal section of the latter substance. Bodybuilders mainly use it enhance fat burning for improved and more noticeable muscle growth. For weight loss, HGH Fragment 176-191 is thought to be considerably more potent than regular growth hormone. It also offers anti-aging benefits as a result of positive effects on IGF-1 levels.
"Paracetamol is used worldwide for its analgesic and antipyretic actions and has been available in Australia since 1956. Caffeine is a stimulant and acts as an analgesic adjuvant, whereby it augments the analgesic effects of pain relievers such as paracetamol. The combination of paracetamol/caffeine (2x500mg/65mg) is indicated for temporary relief of pain and discomfort associated with headaches, tension headaches, osteoarthritis, arthritis, cold and flu symptoms, toothache, dental procedures, muscular aches, sore through and period pain. It also reduces fever.
Technically, it is a “protein-peptide hormone” which means that it consists of 70 amino acids bonded together. Just like the peptides I’ve written about in the past, this means that it must be injected, because otherwise IGF-1 simply degrades in the gut, rendering it useless. Your own human growth hormone release promotes the synthesis of IGF-1 in your liver (and to smaller amounts, synthesis of IGF-1 by your muscles), your liver and muscles then synthesize IGF-1 and then, in the case of your liver, subsequently package the IGF-1 with binding proteins for transport into the blood. In a type of anabolic positive-feedback loop, IGF-1 then further increases growth hormone’s anabolic effects.
Male pattern baldness is genetically switched on at an age that varies very widely (some men in their twenties, some men never – look at the onset in males in your family). When the genetically inherited tendency turns on, then testosterone and it’s stronger (but less concentrated) metabolite, dihydrotestosterone, trigger the slow death of the hair follicles on the scalp in a male baldness pattern.
All relevant GH side effects of numb/tingling hands and arms (especially at night), and water retention will be experienced by the user, but it also has a tremendouse hunger influencing side effect due to its ability to mimic GHRELIN (the hormone that makes our stomach growl and makes us want to eat). Obviously, ravenous hunger isn’t something one would want during a contest diet phase so one might swap from GHRP-6 to GHRP-2, another GH secratagogue which does not make you hungry but which I find is slightly less effective in GH release doses being equal. During the off-season however, hunger can be the bulking bodybuilders’ best friend, so I like to include GHRP-6 solely for this effect in some instances (GH influence aside), in myself and the athletes I help who struggle to find the appetite needed to get through all the food sometimes needed to pack on serious off-season mass.
The Work around is GHRH + GHRP. If GHRH help stimulate growth hormone and GHRP can stimulate twice as much, once you combine the two you are going to increase the amount of growth hormone that can be secreted by 12 times. GHRH will increase the amount of growth hormone that is secreted at the natural times when your body is ready to release the GH. GHRP will selectively send a pulse forcing GH to be secreted. Taking the two together, the GHRP will force a pulse and the GHRH will increase the power of that pulse. Basically, the power of that pulse will be greater, and with more frequency, when using GHRP and GHRH together. This combination gives the option for users to mix and match; thereby, finding which peptides work best for them.
As previously explained, multiple GHRP-6 doses are required throughout the day due to the pulsatile nature of the HGH release, and the administration of these doses are typically administered on average 3 times daily spaced evenly apart. More administrations are acceptable for greater effects on physique and performance, but it is advised that approximately 3 hours in between each injection is ensured so as to allow the pituitary gland to restore its storage of HGH. The most common protocol is as follows:
The impact of the treatment on the neodermal matrix reconstitution was qualitatively graded as described [17, 18]:(0)Immature granulation tissue with a null or incipient formation of collagen fibrils, focally distributed with no alignment and not organized meshwork. Fibrin material prevails in the field. Mallory staining is detected in scarce foci.(1)Scarce collagen fibrils suggestive of a primitive degree of organization, focally distributed, without horizontal alignment along the wound bed. Yet, fibrin occupies more than 50% of the field. Limited number of primitive neoformed vessels with empty lumen. Relative increase of positivity to Mallory staining.(2)A general but coarse image of ECM granulation tissue accumulation, containing intermixed vertically and horizontally oriented collagen fibrils. Full replacement of fibrin by collagen. Fibrin has been fully replaced by collagen. Affinity to Mallory staining is observed.(3)Complete ECM reconstitution, with mature and finely organized collagen fibrils horizontally deposited in the neodermis. The whole matrix appears positive to Mallory staining.
Ironically, it only appears that the version of IGF-1 produced in your own muscle has any true anabolic effects. But nonetheless, many folks who’ve used IGF-1 claim to have experienced significant anabolic effects of injections. However, the only evidence for such anabolic effects have been shown in people who are already clinically deficient in IGF-1.
Years ago, our group examined the cytoprotective effects exerted by the GHRP-6 preventive administration in the hepatic tissue subjected to I/R, as in other distal organs from the ischemic site (ie, lungs, kidneys, and small intestine). Histological and biochemical results allowed us to conclude that the pharmacological preconditioning induced by the GHRP-6 treatment attenuated I/R liver damage. Besides respiratory distress syndrome like pulmonary changes, intestinal transmural infarct and acute tubular necrosis in kidneys were significantly reduced. These results indicated for the first time a systemic cytoprotective effect for the GHRP-6, suggesting its potential efficacy to control the inflammatory response associated with acute I/R and shock, which eventually originated multiple organs damage (MOD). Cytoprotection induced by GHRP-6 treatment was also related to the attenuation in the generation of ROS and preservation of the antioxidant defense reserves. Histological analysis as the assessment of myeloperoxidase activity evidenced a clear anti-inflammatory GHRP-6-induced effect in the liver and remote organs. Moreover, the molecular mechanism mediating the action of GHRP-6 peptide was shown to involve the phosphatidylinositol 3-kinase/RAC-alpha serine/threonine-protein kinase (PI-3K/AKT1) pathway, as the induction of the hypoxia-inducible factor-1 alpha (HIF-1α) all committed in cellular survival.51 Subsequently, Granado et al52 examined the potential anti-inflammatory impact of GHRP-2 in lipopolysaccharide (LPS)-challenged rats. GHRP-2 administration attenuated the effects of LPS on the elevation of circulating levels of transaminases, nitrites/nitrates, and tumor necrosis factor-alpha (TNF-α), via direct interaction with liver nonparenchymal cells. Globally, the exogenous administration of these two synthetic GHRPs appeared to exert a potent hepatoprotective role by attenuating the inflammatory response orchestrated by liver-resident macrophages. Another line of evidences document the benefits of 15-daily injections of GHRP-2 (100 μg/kg) in arthritic rats, so that the treatment ameliorated the external symptoms of arthritis and decreased the circulating levels of interleukin 6 (IL-6) as the nitrite/nitrate release from peritoneal macrophages in vitro. This experiment extrapolated the counter-inflammatory properties of GHRP-2 to a nonepithelial organ and suggested again a direct interaction with ghrelin receptor of immune cells.53 Similarly, effects have been attributed to ghrelin by inhibiting the inflammatory response via AKT1-activated pathway with a concomitant reduction of myeloperoxidase activity, the rate of apoptosis, and oxidative stress.54 All these data suggest that GHRPs exert a mutually inclusive beneficial effect by directly protecting parenchymal organs epithelial cells, and simultaneously by modulating the magnitude of the inflammatory response by direct interaction with the effector immune cells. Supporting the protective effect of GHRP-6 on epithelial organs, a recent study has excellently described and dissected the mechanistic bases on how GHRP-6 prevented gastric mucosal damage induced by water immersion restraint (WRS) and other forms of stress. The data indicated that the protective effect of GHRP-6 on WRS-induced gastric mucosal injury is somehow mediated by peripherally suppressing the vagal efferent effect on the stomach, including gastric acid secretion. Although more studies are clearly demanded, the present findings open the possibility to use GHRP-6 in preventing Curling ulcers.55
I have not used IGF-1 but I have used a stack of Ipamorelin and CJC 1295 no DAC. I did not do any lab tests before, during or after but definitely noticed increased fat loss and better sleep. I was not trying to increase muscle so there was no change to speak of for me. But you are not recommending their use even without IGF-1, is that correct? I do not compete in anything so WADA is not a concern.
Lactating nipples: GHRP-6 side effects include that of increased Prolactin secretion, which can result in lactation from the nipples. This is, however, a rare occurrence but it can happen in sensitive individuals. GHRP-6 exhibits the ability to induce secretion of Cortisol and Prolactin, but studies have shown that the Prolactin and Cortisol increases in most test subjects were not altered at all at GHRP-6 doses of 100mcg or less . Doses above 100mcg are said to increase Prolactin secretion, though minimally, and at these minimal levels lactation should not present itself. However, some users have reported lactation which can be the result of a sensitive individual or the result of much higher doses of GHRP-6. Prolactin can be lowered through the use of a Prolactin antagonist such as Cabergoline, Pramipexole, Bromocriptine, and even vitamin B6.
Increase in ghrelin – Ghrelin is a hormone that the stomach releases when it is empty. It also helps regulate appetite, promotes fat loss in muscle tissue, and helps in healing damaged tendons. There are studies which also point to a direct link between elevated levels of ghrelin and faster repair of tendons. GHRP-6 causes an increase of ghrelin in the body.
As is the case with ghrelin, GHRP 2 has the ability to stimulate secretion of other growth hormones and increase food consumption. When you start taking in GHRP 2 at regular intervals the level of growth hormones being released in your body increases considerably. Another interesting attribute with GHRP 2-based supplements is that they are anti-inflammatory. But the extent of this feature will vary from one person to another as the pituitary somatotrophs of an individual will respond differently to different receptor and so on.
