GHRP-6 is a peptide from the category of growth hormone releasing peptides, ie growth hormone-releasing peptides. The most frequent use of these peptides is an extreme increase in the body's growth hormone production. In June 2011 the Advisory Committee on Medicines Scheduling was referred a proposal by the delegate to consider up-scheduling of five (5) then unscheduled substances contained in cold and cough preparations into Schedule 2. One of these substances was phenylephrine and many public submissions received rejected this proposal on the grounds of the paracetamol/phenylephrine exemptions in the Schedule 2 entry. The committee made similar comments and the delegate agreed that the current exempt from scheduling status of phenylephrine was appropriate.

As an athlete, you can also increase your dosage cycle for a period of 12 to 16 weeks at a time, to maximize your gains. Do so gradually if you opt to go this route. Make sure you increase your daily dosage (1 to 2 doses per day, etc.) gradually. Start off with lower dosage levels as well, and see how it interacts with your body. These studies on human subjects were paralleled by contemporary experimental progresses in basic science, which demonstrated that hexarelin enhanced H9c2 cardiomyocyte proliferation in a dose-dependent manner. Since these were in vitro experiments, they completely excluded a potential intervention of the GH axis and clearly indicated a direct GHRP binding to cardiac cells membranes.32 Weekers et al33 demonstrated that 14 days of pretreatment with GHRP-2, but not GH, selectively protected against the postischemic diastolic dysfunction and myocardial stunning of excised hearts submitted to ischemia/reperfusion in isolated, perfused rabbit hearts.

Nevertheless, we have not elucidated the pathways involved in the GHRP-6-mediated TGFB1 gene expression reduction. Under these circumstances, we have reproducibly observed [7] that GHRP-6 increases PPARG expression which may have counteracted TGF-β1-associated fibrogenic input. The fact that CD36 occupation by GHRP-6 upregulates PPARG gene expression is noteworthy in this context and represents an additional pharmacologic property for this peptide. Although the molecular pathways underlying the antifibrotic effects of PPARγ remain elusive, an antagonistic relationship is proposed between PPARγ and TGF-β1 signaling in fibrosis. For more than a decade ago, PPARγ has been reputed as a fibrosis-response regulating factor and its activation represents an innovative pathway to control fibrotic diseases [31, 32].


Both paracetamol and caffeine are regarded as being well tolerated when used at therapeutic doses and there is a low risk of serious expected or serious unexpected adverse events with these products when taken either alone or in combination. Clinical data demonstrate that paracetamol combined with caffeine significantly out performs paracetamol alone. Paracetamol/caffeine formulations are well established globally. Such formulations are marketed in over 90 countries and have been available unscheduled ranging from 14 years to 25 years. Cumulative post-marketing experience to date with the sponsor’s paracetamol/caffeine combination products is estimated to be in excess of 488 million patients and has revealed no adverse safety signals or reasons for concern with the use of this product in an open sale environment.
A multicenter study comparing the oral GH secretagogue macimorelin with arginine/GHRH found it to be safe, convenient, and of comparable efficacy (82% sensitivity, 92% specificity, and 87% accuracy in diagnosing adult GHD), with a GH cut-off point of 6.8 μg/L for patients with a body mass index (BMI) <30 kg/m2 and 2.7 µg/L for patients with a BMI >30 kg/m2 [268].
GHRP-6 is most commonly provided in small vials of 5 mg, which should be stored under refrigeration. (It is acceptable however for them to be mailed unrefrigerated.) The vial is diluted with a convenient volume of sterile or bacteriostatic water. For example, the vial might be diluted with 2.5 mL of water, yielding a solution of 2 mg/mL (2000 mcg/mL.) After the water addition, the vial again will be stored under refrigeration.
Since CD36 is implicated in angiogenesis regulation, special attention was addressed to the population of neovessels as to their general morphology. By routine staining, we ascertained that GHRP-6 treatment did not reduce the number of vessels, which also exhibited normal structure, organization, and distribution. Furthermore, CD31 expression was detected in all these vascular structures suggesting mature angiogenesis. Conclusively, GHRP-6 administration did not hinder wound angiogenesis in any respect (Figure 3(a)), as compared to placebo-treated wounds (Figure 3(b)). These histological findings support the scoring on the ECM maturation and the quantification of inflammatory cells across the wounds (Table 2).
Though there are a lot of similarities between the two peptides, there is no denying the fact that they also have vast differences. Besides having differences in the release of growth hormones and stimulation of pituitary glands, GHRP 2 is known to have better control in the release of prolactin and Cortisol. That’s not all; GHRP 2-based supplements are relatively cheaper as compared to other supplements that are available in the market, including the ones that are GHRP-6 based. There are a few individuals that opt for GHRP 6 due to the fact that it has lesser impact on prolactin and Cortisol; at least as compared to GHRP 2.
To amend Schedule 2 entry to exempt paracetamol when compounded with caffeine, in a powder or granule product containing 1000mg or less of paracetamol and in tablets or capsules containing 500mg or less of paracetamol when paracetamol is the only therapeutic active constituent and when supplied in primary packs of not more than 20 tablets/caplets or 10 sachets of powders/granules.

For example, one study of women who fed with excess calories over and above their normal metabolic rate noted a 19% increase in IGF-1 after two weeks of overfeeding, with 46% of the weight gain from  lean mass and 54% from bodyfat. Fasting insulin doubled in these women, and testosterone levels also significantly increased.

The discovery of the role of Tβ4 in the process of immune regulation has lead to its use as a valuable therapeutic agent. Tβ4 has been used in the treatment of HIV, AIDS, Influenza, colds, and various infections. It has been utilized in the management of various inflammatory conditions, as well as part of treatment following heart attack due to its cardio and neuroprotective effects.

Additionally and not less relevant, GHRP-6 appears as an excellent partner to combine with other molecules (ie, epidermal growth factor [EGF]) because their exclusive actions seem to achieve a kind of synergism, useful to target the multiples nodes of complex pathophysiological processes, and thus to enhance tissue repair processes.56 Garcia del Barco and coworkers in our group have opened unprecedented avenues, by combining GHRP-6 and EGF as a therapeutic approach to ameliorate the damages of multiple sclerosis,57 peripheral axonal pathology,58 and brain ischemia in animal models.59,60 They have demonstrated that in all these experimental substrates the combined action of GHRP-6 and EGF is associated with a better outcome in both clinical and pathological fields.
The most important initial historical time points in the development of the enlarging ghrelin system were 1973, 1976, 1982, 1984, 1990, 1996, 1998, and 1999 during which the following sequentially occurred: isolation of somatostatin; discovery of unnatural growth-hormone-releasing peptides (GHRPs); isolation of growth-hormone-releasing hormone (GHRH); hypothesis of a new natural GHRP different from GHRH; GHRP+GHRH synergism in humans; discovery of the growth hormone secretagogue GHS/GHRP receptor; cloning of the receptor; isolation; and identification of the new natural endogenous GHRP ghrelin.1

The Work around is GHRH + GHRP. If GHRH help stimulate growth hormone and GHRP can stimulate twice as much, once you combine the two you are going to increase the amount of growth hormone that can be secreted by 12 times. GHRH will increase the amount of growth hormone that is secreted at the natural times when your body is ready to release the GH. GHRP will selectively send a pulse forcing GH to be secreted. Taking the two together, the GHRP will force a pulse and the GHRH will increase the power of that pulse. Basically, the power of that pulse will be greater, and with more frequency, when using GHRP and GHRH together. This combination gives the option for users to mix and match; thereby, finding which peptides work best for them.
In June 2010, the National Drugs and Poisons Schedule Committee (NDPSC) considered the scheduling of paracetamol in combination with ibuprofen. Paracetamol preparations containing 500 mg or less of paracetamol as the only therapeutically active constituent (other than phenylephrine, effervescent agents or guaiphenesin) in packs of 25 or less were exempt from scheduling. However, when these preparations were combined with another therapeutically active ingredient they became Schedule 2. The NDPSC considered that the Schedule 2 entry remained appropriate, but noted the possibility that more robust evidence of additional risk could come to light through any application for product approval with the Therapeutic Goods Administration. The delegate confirmed the NDPSC's decision and the reasons for the decision in August 2010.
Taking into account the broad spectrum of TGF-β1 physiology in the fibroblasts/myofibroblasts differentiation events [33], we deem that the reduction of the local scar cellularity and perichondrial matrix accumulation in those animals receiving GHRP-6 could be attributable to TGFB1 transcriptional and functional switch-off. Since the predominant microscopic aspect of the GHRP-6-treated wounds was characterized by meagre cartilage scars, slimmer perichondrium membranes, and far less active cells, we hypothesize that the peptide somehow attenuates the perichondrial activation response to the trauma and/or a possible mesenchyme-to-mesenchyme redifferentiation process, thus lessening the surge of fibroblast and myofibroblasts. In line with this notion, we had documented that GHRP6 prevented hepatic stellate cells activation by reducing CD68, α-SMA, and vimentin local expressions. All these events could be primarily presided by the GHRP-6-related reduction of TGFB1 and CTGF expression in both parenchymal and nonparenchymal cells [7].
Because some GHRP’s are equally effective as others in increasing GH but differ in effect on hunger or ACTH stimulation, it seems likely that there may be differences in ghrelin receptors between different tissues, or differences in function (for example with cofactors.) This is the most likely explanation for GHRP-6 being effective in stimulating hunger and helping heal tendinitis, while GHRP-2 stimulates hunger less and may have less value for healing.
Similar to other enhancers, it is observed that administrating GHRP-6 along with insulin gets an increased GH response. However, in presence of elevated glucose levels, GHRP-6 does not work well. As a result, consuming carbohydrates or dietary fats before administrating GHRP-6 is a bad idea. Thus the dose should be taken two hours after your last meal and at least thirty minutes before your next meal. Also, GHRP-6 has saturation points. As a result, you want to put a healthy interval between two doses so that your receptors are clear. The best schedule is one dose upon waking up, one post workout, and one before sleeping.
GHRP-6 and all GHRP’s are mimetics of ghrelin, a hormone produced by cells of the stomach in response to a fasted condition, including brief fasts. Ghrelin and ghrelin mimetics work by activating the ghrelin receptor, also called the growth hormone secretagogue receptor (GHS-R1a). Elevated ghrelin levels act towards increasing GH levels by stimulation of ghrelin receptors in the pituitary.

Now these artificially manufactured compounds can replicate certain hormones within the human body to signal or trigger certain effects. In August 2010, the delegate confirmed the decisions of the June 2010 meeting of the NDPSC to transfer leflunomide to Appendix L. Appendix L was a new appendix created to list all of the requirements for dispensing labels previously included in the body of the Poisons Standard (i.e. paragraph 45, Dispensed Medicines, of Part 3, Miscellaneous Regulations) as part of the transitional amendments required to change the Standard for the Uniform Scheduling of Drugs and Poisons No. 24 into the Standard for the Uniform Scheduling of Medicines and Poisons No. 1, under the revised scheduling arrangements commencing 1 July 2010.


Conclusions: Subcutaneous administration of CJC 1295 resulted in sustained, dose-dependent increases in GH and IGF-I levels in healthy adults and was safe and relatively well tolerated, particularly at doses of 30 or 60 ug/ kg. There was evidence of a cumulative effect after multiple doses. These data support the potential utility of CJC 1295 as a therapeutic agent.

Hypertrophic scarring is a form of abnormal, exuberant healing, locally aggressive, and recurrent cutaneous fibroproliferative condition, characterized by excessive extracellular matrix (ECM) accumulation during the cutaneous healing process. Including keloids and hypertrophic scars (HTS), these aberrant processes lead to esthetically disfiguring scars, patients’ psychological stress, and functional impairment [1]. The cellular and molecular mechanisms underlying the formation of these raised dermal scars are poorly understood. Recent whole genome profiling and proteomic studies have led to the identification of regulatory elements with different expression profiles in HTS and keloid tissues [2]. The limited understanding of the pathophysiology of these processes has led to investigating a broad spectrum of potential antihypertrophic scarring candidates [3].

The prescription form of IGF-1 most often injected is “mecasermin”, which goes by the trade name Increlex. Manufactured using recombinant DNA technology, mecasermin is clinically used to treat IGF-1 deficiency and stunted growth. It is also prescribed to patients who have developed antibody resistance to normal growth hormone therapy. Increlex is actually identical to natural IGF-1, meaning that it has the identical 70 amino acid sequence of IGF-1 that the body produces. In other words, it’s not some kind of growth hormone “precursor”. It’s just straight up IGF-1.
But for maintenance of adequate and natural IGF-1 and growth hormone, and to achieve that sweet spot of not becoming to pro-growth while also not becoming a weak, muscle-less noodle, that sweet spot of producing adequate insulin without producing too much, and that sweet spot of increasing cellular repair without letting cellular division get “out of control”, I have indeed been implementing three specific strategies: my IGF-1 “trilogy”.

This is caused by an IGF-1-induced drop in blood phosphate levels, and in the bodybuilding community is often prevented by administering phosphate with the IGF-1.
More than a decade ago, CD36 was identified as one of the GHRP-6 receptors [12]. This is a scavenger receptor endowed with multiligand and multifunctional capabilities and is expressed by a broad constellation of mammalian cells [13]. Granulation tissue neovascularization is perhaps the most renowned physiological role of CD36 in wound healing [14]. Serendipitous observations of our laboratory indicated that CD36 mRNA transcript appeared abundantly represented in clinical samples of granulation tissue of either acute (deep burn injuries) or chronic (pressure ulcers) wounds, as in laboratory rat’s controlled full-thickness wounds. This finding incited us to speculate on the effects associated with CD36 agonistic stimulation beyond that of the angiostatic action via thrombospondin binding [15]. Here we provide the first experimental evidence on the favorable impact of the topical administration of GHRP-6, as a candidate to qualitatively improve the healing process.
Then there’s colostrum. Colostrum is packed with growth factors, including IGF-1, that amplify lean muscle gains and increase the body’s ability to burn fat. In many studies, colostrum has been shown to restore IGF-1 and stimulate IGF-1 production. Colostrum is also a natural immunity drug, containing antibodies and antigens that knock out disease-causing agents such as bacteria, viruses, and fungi.
In a study designed to assess the effect of both the estrogen and GHRP-6 on the cardiovascular and metabolic diseases in ovariectomized (OVX) rats, Elbassuoni, et al found that although GHRP-6 failed to produce significant change in body weight gain and food intake, it clearly reversed the effect of OVX on fasting serum glucose, insulin, insulin resistance, and the assessed lipid fractions. They concluded that the effect of GHRP-6 on improving dyslipidemia after OVX was even more potent than that of estrogen.12 Furthermore, the mechanism of action of GHRP-6 has been more extensively studied in experimental models with obese subjects, and was shown to be a powerful GH releaser in obesity, and to release GH independently of the hypothalamic factors (GHRH and somatostatin).13
Finally, patients deficient in growth hormone who get IGF-1 injections have shown increased rates of fat loss and fat oxidation. One theory for this is that, as you’ve just learned, IGF-1 can suppress circulating insulin, which would allow more burning of fatty acids from fat cells. This makes sense, since we do know that fat cells contain IGF-1 receptors, and this means that IGF-1 can interact with fat cells.
Researchers around the globe suggest that the effectiveness of growth hormones depends a lot on the physical condition of the subject being administered with the drug. If the subject is obese, then there may not be the desired level of hormone secretion. Obesity seems to affect the effectiveness of GHRP-6 but if the subjects are not obese, the effects of this drug is likely to be the same for all gender or age groups, subject to the administered dosage.
Mostly, these peptides are sold as lyophilized powder in 2mg containers. Bacteriostatic water should be mixed with the powder in order to reconstitute it. To make the dosage of 100mcg per injection, 2ml bacteriostatic water should be mixed into 2mg of lyophilized powder. This reconstituted mixture should be then injected inside the muscles or under the skin. The mixture should be kept under refrigeration at all times otherwise it will degenerate and will not be effective anymore.
In addition, scientists have observed that those who supplemented with IGF-1 experienced a preponderance of new brain cell growth and new muscle mass and new studies confirmed this to be true, even among individuals with both brain damage and muscle-wasting sarcopenia. Furthermore, research studies have found IGF-1 to increase feelings of youthfulness, improve well-being, and even to alleviate depression.

Another very positive benefit of CJC1295 is its ability to promote slow wave sleep. Slow wave sleep (SWS) is also known as deep sleep and is the portion of sleep responsible for the highest level of muscle growth and memory retention. SWS is decreased significantly in older adults and also with people who tend to exercise later in the evening.  This peptide has a benefit to side effect ratio that exceeds all others currently being legally sold and would make a great addition to ones training regimen or post cycle therapy.
The experiment in rats, based on clean full-thickness controlled wounds, indicated that GHRP-6 pharmacodynamics has likely involved attenuation of immunoinflammatory mediators, their effector cells, and the reduction of fibrosis-inducing cytokines. The concerted action of these two elemental mechanisms may have theoretically translated into a particular modulation of fibroblasts response to injury, leading to precocious closure with a reduced scarring. Outstandingly, the mechanisms underlying this pattern of healing do not appear to interfere with the angiogenic repopulation nor with the reepithelialization process.

The locally secreted pro-inflammatory cytokines are involved in a self-perpetuating process in the ROS chain reaction, inflammation, and cellular damage.
