White male New Zealand rabbits (4.3–4.5 kg) were used in four independent and extemporaneous experiments. Three to four wounds were created on the ventral side of each ear, down to the surface of the cartilage, using a 6 mm diameter punch biotome (Acuderm) as described . For the surgical procedures, rabbits were anesthetized with intramuscular ketamine (60 mg/kg) and xylazine (5 mg/kg). In order to ensure an exuberant scarring, the perichondrium was carefully scrapped with the surgical blade. The wounds were made on each side of the midline, avoiding the central ear artery and the marginal ear veins. In three experiments, rabbits were randomly assigned to either GHRP-6 (400 μg/mL) treatment or 1% CMC placebo gel. The jelly solutions were administered using 1 mL sterile disposable syringes; 250 μL was applied to each wound, which for the group of GHRP-6 represented an actual dose of 100 μg per wound. Treatments were initiated immediately after surgery and continued thereafter until day 30, when most of the wounds had already completed reepithelialization.
The wounds were monitored and followed from day 14 until day 30 after wounding so as to detect the nodular firm consistency that precedes the clinical exuberance. The animals remained in observation for another 20 days after GHRP-6 administration had been completed. The incidence of firm, protruded nodules with nipple-like appearance arising in resurfaced wounds was registered weekly until day 50. After euthanasia (anesthesia overdose), the samples were collected in block, longitudinally bisected along the largest point of nodular growth. One hemisection was nitrogen frozen for additional studies and the other one was fixed in 10% neutral buffered formaldehyde and processed for histology. Five-micrometer sections were stained with H/E staining. Scar overgrowth was measured using the previously described scar elevation index (SEI) based on the cross-sectional scar area to the area of tissue excised to induce the wound . Blinded researchers measured the sections using the ImageJ software package, version 1.46r.
Another major difference between GHRP 2 and GHRP 6 is that the former is a bit stronger and releases a lot more Growth Hormone as compared to GHRP 6. Dosing will ordinarily be at least twice per day and preferably 3x/day for best effect, taken at least 30-60 minutes before a meal and at a time of non-elevated blood sugar (in other words, after blood sugar has had time to fall since the most recent meal.) In 1999, seven adult patients with GH deficiency and LV failure received hexarelin administrations. The GH response to hexarelin was negligible in these patients. Moreover, hexarelin administration increased their left ventricular ejection fraction (LVEF) without changing catecholamine levels, mean blood pressure (MBP), or cardiac output. For the first time, the acute administration of hexarelin proved to induce a positive inotropic effect in humans, which is GH independent and mediated by specific myocardial receptors for a GH secretagogue peptide.29 A subsequent study involving hexarelin administration to normal adults, severe GH-deficient patients (N = 7), and patients with severe ischemic DCM (N = 12) confirmed that the acute administration of hexarelin exerts a GH-independent positive inotropic effect likely mediated by specific GHRP myocardial receptors.30 This pioneering group subsequently evaluated the cardiac performances of the acute hexarelin administration (2.0 µg/kg, i.v.) in patients undergoing bypass surgery in comparison to patients given GH-releasing hormone, recombinant human GH, or placebo. The study concluded that the acute administration of hexarelin improved cardiac performance without any relevant variation in systemic vascular resistance and induced a reduction of wedge pressure and, significantly, that these cardiotropic effects were not shown by the other concurrent interventions.31
Because some GHRP’s are equally effective as others in increasing GH but differ in effect on hunger or ACTH stimulation, it seems likely that there may be differences in ghrelin receptors between different tissues, or differences in function (for example with cofactors.) This is the most likely explanation for GHRP-6 being effective in stimulating hunger and helping heal tendinitis, while GHRP-2 stimulates hunger less and may have less value for healing.

Conclusions: Subcutaneous administration of CJC 1295 resulted in sustained, dose-dependent increases in GH and IGF-I levels in healthy adults and was safe and relatively well tolerated, particularly at doses of 30 or 60 ug/ kg. There was evidence of a cumulative effect after multiple doses. These data support the potential utility of CJC 1295 as a therapeutic agent.

More than a decade ago, CD36 was identified as one of the GHRP-6 receptors . This is a scavenger receptor endowed with multiligand and multifunctional capabilities and is expressed by a broad constellation of mammalian cells . Granulation tissue neovascularization is perhaps the most renowned physiological role of CD36 in wound healing . Serendipitous observations of our laboratory indicated that CD36 mRNA transcript appeared abundantly represented in clinical samples of granulation tissue of either acute (deep burn injuries) or chronic (pressure ulcers) wounds, as in laboratory rat’s controlled full-thickness wounds. This finding incited us to speculate on the effects associated with CD36 agonistic stimulation beyond that of the angiostatic action via thrombospondin binding . Here we provide the first experimental evidence on the favorable impact of the topical administration of GHRP-6, as a candidate to qualitatively improve the healing process.

During the last 15 years, a plethora of experimental evidence supports the pharmacological benefits of the exogenous administration of synthetic growth hormone-releasing peptides (GHRPs). In parallel to their growth hormone-releasing action, these agents exert cytoprotective effects encompassing cardiac and extracardiac organs . GHRP-6 is a class of peptidyl GH secretagogue, similar to met-enkephalin, that has reproducibly shown antinecrogenic and antiapoptotic properties in multiple experimental scenarios, including ischemia/reperfusion [9–11]. Globally speaking, exogenously administered GHRP-6 has broadly been shown to act as a prosurvival factor for cells and tissues threatened by otherwise lethal insults.
According to pilot studies, our group determined that 400 μg/mL represented an optimal dose level by reducing inflammation, promoting collagen fibers alignment, while aborting the onset of HTS in rabbit ears. A lower dose (200 μg/mL) did not prevent the exuberant phenotype whereas a higher dose (800 μg/mL) delayed reepithelialization in rats and rabbits (data not shown).
In no particular order of importance, here they are: I swallow colostrum capsules every morning, I drink raw animal milk such as camel milk and goat milk in moderation, and I use the equivalent of around 30 grams of grass-fed whey protein each day in a smoothie (if you’re vegan or if whey protein doesn’t agree with your stomach, you can combine digestive enzymes with a vegan protein such as brown rice protein, pea protein or hemp protein for an effect similar to whey protein).
Additionally and not less relevant, GHRP-6 appears as an excellent partner to combine with other molecules (ie, epidermal growth factor [EGF]) because their exclusive actions seem to achieve a kind of synergism, useful to target the multiples nodes of complex pathophysiological processes, and thus to enhance tissue repair processes.56 Garcia del Barco and coworkers in our group have opened unprecedented avenues, by combining GHRP-6 and EGF as a therapeutic approach to ameliorate the damages of multiple sclerosis,57 peripheral axonal pathology,58 and brain ischemia in animal models.59,60 They have demonstrated that in all these experimental substrates the combined action of GHRP-6 and EGF is associated with a better outcome in both clinical and pathological fields.
The use of peptides is not necessarily a case of one or the other. While you can derive benefit from using either GHRH or GHRP, the effects can be amplified by using both. This, for one, will help to better keep levels of somatostatin under control. The amount of that hormone is an issue you will be forced to contend with when using only GHRH. This is because it is known to inhibit growth hormone.
Users get even greater growth hormone release from this peptide than both GHRP-6 and GHRP-2. Much like other GHRP and GHRH peptides, it is believed to be useful for muscle building, fat loss and anti-aging. Hexarelin is stronger than practically all other growth hormone releasing peptides at all dosing levels. But this means it has higher potential to desensitize, regardless of dose or whether breaks are observed.
Scheduling both enobosarm and SARMs would address the potential problem of misuse and abuse. The class entry for SARMs was recommended as there are other SARMs being developed. Patients being treated with these drugs would require medical diagnosis, monitoring and management. There is access to SARMs that are more toxic than enobosarm. If only one SARM was scheduled, consumers would be able to source another SARM.
In 1982, the natural hormone "Growth Hormone Releasing Hormone" (GHRH) was identified after a prolonged search. Soon, researchers discovered that those GH-Releasing Peptides (specifically GHRP-6 & GHRP-2) followed a mode of action which bound them to and was mediated through receptors different from those for GHRH. Furthermore, researches discovered that these GH-Releasing Peptides acted synergistically with the natural hormone Growth Hormone Releasing Hormone (GHRH), which is related to Sermorelin, in both laboratory animals and humans to produce large releases of Growth Hormone. In the 1980s, the first highly potent GH-Releasing peptide, GHRP-6, was developed. Due to a strong GH release response from the the peptide, it became the first member of a class called Growth Hormone secretagogues. GHRP-6 is a hexapeptide composed of 6 amino acids: L-Histidine, D-Tryptophan, L-Alanine, L-Tryptophan, D-Phenylalanine and L-Lysine. The "L" form of an amino acid is the naturally occurring form and often in the nomenclature the "L" is dropped. The "D" form does not occur in nature and is the isomeric form (i.e. mirror image) of the naturally occurring "L" form. GHRP-6 (His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) is composed of both natural and isomeric forms of those 6 amino acids.This sequence provides a signal to the body to begin secreting Growth Hormone release while also blocking Somatostatin, a hormone that inhibits the release of Growth Hormone.
I stopped the colostrum and my ” symptoms ” subsided, seems I have a moderately enlarged prostate which doesn’t run in my family on either side, my question is could the colostrum possibly cause the prostate to enlarge due to the igf-1 at a certain age,? due to a possible decline in testosterone, or could the benefits of colostrum outweigh the prostate issue?
Since CD36 is implicated in angiogenesis regulation, special attention was addressed to the population of neovessels as to their general morphology. By routine staining, we ascertained that GHRP-6 treatment did not reduce the number of vessels, which also exhibited normal structure, organization, and distribution. Furthermore, CD31 expression was detected in all these vascular structures suggesting mature angiogenesis. Conclusively, GHRP-6 administration did not hinder wound angiogenesis in any respect (Figure 3(a)), as compared to placebo-treated wounds (Figure 3(b)). These histological findings support the scoring on the ECM maturation and the quantification of inflammatory cells across the wounds (Table 2).
In 2005, we undertook a porcine model of AMI via left circumflex artery occlusion for 1 hour followed by a 72-hour reperfusion period. GHRP-6 rescued ischemic myocardium from death for over 70% of the area at risk (Figure 3), and that in addition to enhance survival signaling pathways/gene expression of the PI-3K, AKT1, and BCL2 pathways, GHRP-6 decreased reactive oxygen species (ROS) spillover, the inflammatory marker CRP, and preserved the antioxidant defenses.45 These antioxidant and anti-inflammatory properties have also been attributed to GHRP-2 when its antiatherogenic potential was examined in ApoE(−/−) mice so that 12/15-lipoxygenase, interferon gamma, and macrophage migration inhibitory factor (MMF) gene expression were accounted. Furthermore, in cultured aortic smooth muscle cells, GHRP-2 prevented the generation of peroxides, the downregulation of IGF-1 receptor, and the commitment of apoptosis.46
All relevant GH side effects of numb/tingling hands and arms (especially at night), and water retention will be experienced by the user, but it also has a tremendouse hunger influencing side effect due to its ability to mimic GHRELIN (the hormone that makes our stomach growl and makes us want to eat). Obviously, ravenous hunger isn’t something one would want during a contest diet phase so one might swap from GHRP-6 to GHRP-2, another GH secratagogue which does not make you hungry but which I find is slightly less effective in GH release doses being equal. During the off-season however, hunger can be the bulking bodybuilders’ best friend, so I like to include GHRP-6 solely for this effect in some instances (GH influence aside), in myself and the athletes I help who struggle to find the appetite needed to get through all the food sometimes needed to pack on serious off-season mass.
In 1984, a synthetic hexapeptide, His-d-Trp-Ala-Trp-d-Phe-Lys-NH2 (GHRP-6), was identified by Bowers and colleagues. This hexapeptide was shown to potently stimulate GH release in vitro and in vivo by an unknown mechanism. Because of its poor oral bioavailability (0.3%) and short half-life (20 min) in human serum, GHRP-6 was selected only as a structural model to design a nonpeptide mimetic. Based on the structure–activity relationships (SARs) of GHRP-6, the nonpeptidyl growth hormone secretagogue (GHS) L-692,429 was identified by Smith et al. in 1993. This nonpeptidyl GHS synergizes with GHRP-6 to stimulate GH release and cAMP production, accompanied by a significant increase in intracellular calcium concentration ([Ca2 +]i), indicating that this nonpeptidyl GHS acts through a distinct signal transduction pathway. In 1995, a potent oral GHS L-163,191 (MK-0677) was reported by Patchett et al. This agent was found to have excellent oral bioavailability and specificity in its release of GH, without significant effect on plasma levels of other hormones such as aldosterone, luteinizing hormone, thyroxine, and prolactin.
Similar to GHRP 2, this peptide is a more potent releaser of growth hormone, also acting on the ghrelin receptors of the anterior pituitary. Also like GHRP 2, GHRP 6 leads to increased growth hormone production, increased lead body mass, and decreased adiposity. Due to the peptide’s ghrelin-like properties, administration can lead to increased appetite.
