GHRP mechanism of action. GHRPs are endowed with the ability to bind two different receptors that seem to mediate its cytoprotective and other pharmacological properties (GHS-R1a and CD36). The main biological properties/pharmacological actions of GHRP-6 as cyto- and cardioprotective candidates are summarized as follows: Inotropic: mediated by an elevation of Ca2+ influx via PLC/DAG/PKC, through the voltage-gated calcium channel, triggering Ca2+ release from thapsigargin-sensitive intracellular stores, which translated in a positive inotropic response without a chronotropic effect. Anti-fibrotic: via upregulation of PPARγ, which is followed by a transforming growth factor-beta (TGF-β), CTGF, and platelet-derived growth factor (PDGF) downregulation. Anti-inflammatory: blunts NFκB expression and activation. Cell survival: it involves the phosphatidylinositol 3-kinase/RAC-alpha serine/threonine-protein kinase (PI-3K/AKT1) pathway, as the induction of the hypoxia-inducible factor-1 alpha (HIF-1α). Cardioprotective: as shown, it involves different biological actions that converge to enhance cardiomyocytes survival. Vasodilatory: it seems to involve e-NOS upregulation and endothelin activity reduction. Anabolic: it is mediated by the IGF-1/AKT1 and mTOR pathway activity.
For those who are just getting started, make sure you go gradually. Start off with an eight-week cycle, and start off with 200 mcg (rather than 300) per day. Doing a test run will allow you to see how your body is going to react. If all goes well, you can then increase your dosage cycle to an 8 to 12 week period, and possibly add an additional injection dose daily, or increase to 300 mcg with each use.

Activation of CD36 in perfused hearts by hexarelin was shown to increase coronary perfusion pressure in a dose-dependent manner. Contrariwise, this effect was lacking in hearts from CD36-null mice and hearts from spontaneous hypertensive rats genetically deficient in CD36.19,20 Thus, it is currently accepted that two cardiac receptor subtypes mediate the pharmacological actions of GHRP-6, GHRP-2, and hexarelin.21,22
TO-2 hamster model of DCM was characterized by progressive LV dilation, LV wall thinning, LV systolic dysfunction, and reduced life span; both GHRP-2 and GHRP-6 ameliorated all the dysfunctional ventricular parameters and reduced the progression of the DCM.34 We also examined the potential impact of GHRP-6 in a rat model of DCM/heart failure induced by doxorubicin (DX). The concurrent administration of GHRP-6 was undertaken with the purpose to study the potential prophylactic impact before the cardiac function demise. As part of the prolonged treatment with DX, the concurrent administration of GHRP-6 completely prevented failure of cardiac function, which was evaluated as the percentage of ejection fraction by echocardiography (Figure 2, prevention). This effect significantly increased the survival of animals. Similar results were obtained in the therapeutic administration schedule, with functional recovery of cardiac muscle to physiological levels (Figure 2, regression), also attenuating systemic damages and, consequently, decreasing the mortality rates of rats. In the experimental model of DX-induced cardiac and systemic damage, GHRP-6 additionally attenuated various extracardiac damages observed in the renal tubular and bronchoalveolar epithelial structures as in the hepatic parenchyma.35

Ghrelin has been linked to inducing appetite and feeding behaviors. Circulating ghrelin levels are the highest right before a meal and the lowest right after. Injections of ghrelin in both humans and rats have been shown to increase food intake in a dose dependent manner. So the more ghrelin that is injected the more food that is consumed. However, ghrelin does not increase meal size, only meal number. Ghrelin injections also increase an animals’ motivation to seek out food, behaviors including increased sniffing, foraging for food, and hoarding food. Ghrelin also readies the body for the incoming nutrients by stimulating gastrointestinal motility and gastric acid secretions.

The prescription form of IGF-1 most often injected is “mecasermin”, which goes by the trade name Increlex. Manufactured using recombinant DNA technology, mecasermin is clinically used to treat IGF-1 deficiency and stunted growth. It is also prescribed to patients who have developed antibody resistance to normal growth hormone therapy. Increlex is actually identical to natural IGF-1, meaning that it has the identical 70 amino acid sequence of IGF-1 that the body produces. In other words, it’s not some kind of growth hormone “precursor”. It’s just straight up IGF-1.
GHRP-6 is normally always manufactured as lyophilized (freeze-dried) powder contained in vials in amounts of 5mg. Some companies might manufacture amounts greater or lesser than 5mg per vial, but the standard is generally 5mg/vial. The lyophilized powder contained within the vial will need to be reconstituted with bacteriostatic water in order for it to be injected. After reconstitution, the solution must then be refrigerated in storage. If left in hot environments or in room temperature environments for extended periods of time, the protein structure will degrade and become ineffective. For reconstitution, users will typically mix 3ml of bacteriostatic water with the powder gently. However, users can and do frequently reconstitute the powder with less (or more) water which will yield different concentrations of GHRP-6. For example, reconstitution of 5mg of powder with 3ml of water will yield GHRP-6 doses of 166mcg per 0.1ml (or 10iu on an insulin syringe).
Prolactin, or estrogen gynecomastia issues: Those that are sensitive to prolactin based gynecomastia are open to problems with GHRP-6. Luckily, this is a rare occurrence, and it can be easily combated with dostinex. However, those who stack this compound with anabolic androgenic steroids may have a greater chance of getting gynecomastia. Thats why an AI (aromatase inhibitor) should be used during this kind of cycles.
For example, if 100mcg more were to be administered after the first 100mcg (making the effective dose of 200mcg), then the second dose will achieve only 50% of what the first dose already did. A 100mcg more (making a total of 300mcg) will achieve only 25% more of the initial dose. This implies that, in order to increase the effect of the compound, only a little more of it can be successfully administered after the saturation dose.

However, there is always a but, if you take more than 150mcg the effects of the gastric emptying can be so strong that you may have the urge to severely stuff yourself with food, so if you're on a bulking cycle this is a great side effect, and, considering the price, it's a very cost-effective one. Therefore, during a bulking run, I rate this as the number one aid in increasing appetite, as you also get very a good anabolic effect and increased strength.
Ghrelin has many activities in the body besides stimulating GH release. It stimulates appetite, is cardioprotective, can help protect cells against oxidative damage, can reduce inflammation and promote healing, and can promote fat-burning in muscle. There is also some effect on increase in cortisol production via increase in ACTH, and increase in prolactin. However, where the activity of ghrelin is comparable to that which ordinarily occurs during fasting, effects on cortisol and prolactin likewise are comparably only to that experienced while fasting.
The authorities have branded it as a banned substance in the competitive athletics and bodybuilding to prevent the unfair advantage users are likely to gain from this drug. But many athletes and bodybuilders continue to pursue the drug actively. Then there are those people who are ready to try just about anything to lose all of their extra weight. There is always heavy demand for fat cutters and the demand for GHRP-6 is no exception. A majority of the demand groups don't have prescriptions for this drug and are likely to procure this from the black market or the internet. This is where the authorities need to step in.
You’re no doubt taking it for the fairy tale positive side effects, which have already been outlined, but like any caper about something enchanted, the magic comes with a price. For GHRP6 these can include flu-like symptoms, joint aches, headaches and water retention. Prolonged use can give you a tingling feeling in your skin than can also lead to a loss of sensitivity to touch. Yeah, you don’t want it down there. Fortunately, this is often in rare cases and when you consider even garden-variety paracetamol can dish out hives, diarrhoea and nausea then by comparison these aren’t huge risk factors. The biggest drawback is that it has a meagre half-life of 15-60 minutes, which means you have to take it daily for it to be effective, with the primary method of administration being a big ole fat needle. So the idea of turning the glutes into something that resembles nanna’s pincushion may deter pretty much all-conscientious pain objectors.
