The effect of GHSs on GH release is dose dependent and more reproducible than that of GHRH. Although this signaling and activation cascade has not been described for myocardial cells so far, the potential existence of these phosphorylative and mitochondriogenic mechanisms in the heart, and its potential amplification by GHRP ligands, may eventually contribute to myocardial salvage during critical ischemia periods.47 In a more recent study based on a myocardial infarction model, and addressed to examine whether hexarelin treatment can compensate for ghrelin deficiency in ghrelin-knockout mice, the mortality within two weeks was significantly lower in the hexarelin (6.7%) and ghrelin groups (14.3%) than in the vehicle group (50%). Furthermore, hexarelin was more effective than ghrelin as judged by the ejection fraction and other LV-dependent physiological constants as dP/dt max and dP/dt min, which is a measure of LV global contractility.48

Growth Hormone Releasing Peptides (GHRP) are a class of compounds, which stimulate the release of growth hormone. I have not used IGF-1 but I have used a stack of Ipamorelin and CJC 1295 no DAC. I did not do any lab tests before, during or after but definitely noticed increased fat loss and better sleep. I was not trying to increase muscle so there was no change to speak of for me. But you are not recommending their use even without IGF-1, is that correct? I do not compete in anything so WADA is not a concern.

Although the history of some of the foremost biomedical discoveries is permeated by serendipity,4 we deem that the well-established pivotal role of the GH/insulin-like growth factor-1 (IGF-1) axis for cardiomyocyte physiology, and the subtle alterations of this axis within the pathogenicity of dilated cardiomyopathy (DCM) and left ventricular (LV) dysfunction, ignited the idea of assessing the potentiality of GHRP to alleviate cardiac pathologies.5 It was far to be anticipated on those early days, however, that the GHRP-mediated cardiotropic and cytoprotective effects are superior to those shown by the exogenous administration of GH and are not shared by GH-releasing hormone (GHRH) and that, importantly, GHRPs exert their pharmacological actions via GH-independent pathways that obviously represented another turning point in this history.3


But IGF-1 injections may soon be a thing of the past. Future use of IGF-1 will no doubt involve gene therapy, which directly targets genes that produce IGF-1 in muscle, usually by attaching specific gene activators to an inactive virus or vector that then enters into muscle cells. Studies in mice show that a procedure like this can cause  a 15% increase in muscle mass, along with a 14% increase in strength. Gene therapy in old mice has been shown to cause to a 27% increase in strength, along with regeneration of aging muscle. In one mouse study, the IGF-1 gene was placed in the animals’ glutes and calves, which resulted in up to a 115% increase in muscle-cross-sectional area.
Ipamorelin is very similar to the growth hormone releasing peptides (GHRPs) GHRP 2 and GHRP 6 in that it mimics ghrelin (the hunger hormone) and targets a specific HGH pulse. However, unlike other GHRPs, this peptide doesn’t affect the release of cortisol, acetylcholine, prolactin and aldosterone thereby minimizing side effects experienced with other GH therapies, such as increased hunger. Because there are virtually no negative side effects, Ipamorelin can be prescribed more aggressively and more frequently than other therapies without the risk of elevated cortisol and acetylcholine blood plasma levels. Extreme hunger stimulation: It has been previously mentioned in this profile already that almost all GHRPs will stimulate hunger simply by virtue of the fact that they are Ghrelin mimetics (they mimic the action of the hormone Ghrelin in the body), but GHRP-6 has demonstrated both anecdotally as well as clinically to stimulate the largest hunger increases in comparison to all other GHRPs[2]. GHRP-6 tends to stimulate what can only be described as agonizing hunger, and is commonly misconceived as a hypoglycemic episode. The hunger resultant from GHRP-6 is, in fact, simply the peptide acting on Ghrelin receptors that signal hunger to various regions in the brain. No studies or any anecdotal evidence demonstrates hypoglycemic blood glucose readouts following GHRP-6 administration. Following a meal, the hunger should subside. Many users have also reported the intensity of the hunger diminishing several weeks into GHRP-6 cycles but not completely disappearing.

The delegates have decided to accept the advice of the expert advisory committees that the current scheduling of preparations in Schedules 5 and 6 remains appropriate. The delegates noted the decision to retain listing of teeth whitening preparations containing up to 6 per cent hydrogen peroxide or 18 per cent carbamide peroxide in Schedule 5, or exempt when below 3 per cent and 9 per cent respectively, is to some extent also consistent with the evaluation of such products by the European Commission Scientific Committee on Cosmetics and Non Food Products intended for Consumers (SCCNFP).
Ryan also brings up a good point to bring up the fact that GHRH receptors are typically not desensitized with higher dosing but again not much accomplished with CJC-1295, ,MOD GRF 1-29 where we know the 100mcg is a saturation dose. on the other hand we can desensitize the GHRP receptor if we start increasing the dosing significantly above the saturation dosing  of 100mcg. I think you are fine at a 150 mcg dosing but again how much more benefit are you really getting.

I have questions about combined therapy of CJC 1295 and Ipamorelin at the same time on a daily basis for both. The compounding pharmacies do not clearly state whether the CJC is with or without DAC. If it is the CJC with DAC, which sustains elevated GH and IGF-1 for several days, would taking it nightly in conjunction with the Ipramorelin, that is suggested to be taken TID but is being recommended only once at night, be over-stimulatory? If the CJC is without DAC, why take two pepetides simultaneously ,that have similar effects? I am just not clear why taking a daily dose of CJC with Ipamorelin as a single dose is better than taking the CJC with DAC twice per week alone or take the CJC with DAC for a while then switch to the Ipamorelin for a while?
Growth Hormone Releasing Peptide-6 is responsible for releasing growth hormone in appropriate quantities in the body and it does so by stimulating the pituitary gland. In more recent years, these data were further substantiated using again the TO-2 hamster DCM biomodel in which GHRP-2 reduced the progression of LV remodeling, dysfunction, and the ensued myocardial fibrosis by an antioxidant mechanism.36 The abovementioned myocardial fibrotic process amelioration reveals an additional potential use for GHRP in an unmet medical need. Chronic treatment with hexarelin in spontaneously hypertensive rats, in addition to decreasing ventricular hypertrophy, diastolic dysfunction, and high blood pressure, significantly reduced cardiac fibrosis by decreasing interstitial and perivascular myocardial collagen deposition and myocardial hydroxyproline content. Mechanistically, hexarelin treatment increased matrix metalloproteinase (MMP)-2 and MMP-9 activities and decreased myocardial mRNA expression of tissue inhibitor of metalloproteinase (TIMP)-1.37

Results: After a single injection of CJC 1295, there were dose dependent increases in mean plasma GH concentrations by 2- to 10-fold for 6 d or more and in mean plasma IGF-I concentrations by 1.5- to 3-fold for 9–11 d. The estimated half-life of CJC 1295 was 5.8–8.1 d. After multiple CJC 1295 doses, mean IGF-I levels remained above baseline for up to 28 d. No serious adverse reactions were reported.

Effect of GHRP-6 on the left ventricular ejection fraction in an experimental model of dilated cardiomyopathy (DCM). Echocardiography data were derived from our DCM model including the prevention and the regression study protocols in rats.35 The prevention protocol conceived the concurrent administration of GHRP-6 as part of a prolonged treatment with doxorubicin. The concomitant GHRP-6 completely prevented cardiac function failure evaluated as the percentage of ejection fraction by echocardiography. The regression approach examined the GHRP-6 intervention once LVEF was already deteriorated. As shown, the therapeutic administration schedule introduced a full functional recovery of cardiac muscle. GHRPs are not simply surrogates of GHRH, instead GHRP-6 is an artificial activator of a separate newly discovered receptor called Growth Hormone Secretagogue Receptor (GHS-R). Soon Ghrelin was discovered, the endogenous ligand that binds to the GHS-R. Both Ghrelin and all the synthetic compounds such as GHRP-6 were termed "Growth Hormone Secretagogues" (GHSs). The wounds were monitored and followed from day 14 until day 30 after wounding so as to detect the nodular firm consistency that precedes the clinical exuberance. The animals remained in observation for another 20 days after GHRP-6 administration had been completed. The incidence of firm, protruded nodules with nipple-like appearance arising in resurfaced wounds was registered weekly until day 50. After euthanasia (anesthesia overdose), the samples were collected in block, longitudinally bisected along the largest point of nodular growth. One hemisection was nitrogen frozen for additional studies and the other one was fixed in 10% neutral buffered formaldehyde and processed for histology. Five-micrometer sections were stained with H/E staining. Scar overgrowth was measured using the previously described scar elevation index (SEI) based on the cross-sectional scar area to the area of tissue excised to induce the wound [21]. Blinded researchers measured the sections using the ImageJ software package, version 1.46r.

Drug discovery is an uncertain ground in which disappointments and rewards are encountered. Most of those who have been involved in GHRP research have enjoyed clear-cut data, which in most of the cases are all in with very few outs. Exceptionally, a pharmacologically active agent appears to be endowed with such a variety of useful properties as to make it highly drugable. The fact that synthetic GHRPs bind at least two different and biologically significant receptors that seem not to be redundant in nature and are largely represented in most organs and tissues broadens their biological activities and increases their pharmacological potentialities. This suggests that GHRPs may stimulate multiple cells and simultaneously trigger different signaling pathways. The information gathered so far in terms of the molecular cytoprotective mechanism of GHRPs is inconclusive and fragmentary, which has become difficult to disclose the hidden facts behind their biological effects. Nevertheless, it is reasonable that these molecules share the ability to knock life-sensitive pathways and restore critical organelle physiology at very proximal levels. Beyond their ability to enhance the survival of a diversity of cells and tissues before adverse episodes, GHRP members exert an agonistic effect of the GH/IGF-1 axis, promoting anabolia and deterring catabolism and sarcopenia.

Aside from the limitations of this work to fully elucidate the underlying mechanism by which GHRP-6 mediated the refinement of the wounds fibrogenesis in the rats experiment, an important contribution is the unprecedented evidence that the peptide reduced the onset of HTS in the rabbit’s ear model. This represents an extension of the GHRP-6 antifibrotic potential demonstrated years ago by our group in an animal model of liver fibrosis [7]. Nevertheless, and in contrast to the liver fibrosis data, we have no evidence that GHRP-6 is able to revert the consolidated HTS following repeated experimental attempts. Thus, the reproducible findings regarding GHRP-6-mediated HTS prevention are based on the immediate and consecutive administration of the molecule once the injury is induced.
Though there are a lot of similarities between the two peptides, there is no denying the fact that they also have vast differences. Besides having differences in the release of growth hormones and stimulation of pituitary glands, GHRP 2 is known to have better control in the release of prolactin and Cortisol. That’s not all; GHRP 2-based supplements are relatively cheaper as compared to other supplements that are available in the market, including the ones that are GHRP-6 based. There are a few individuals that opt for GHRP 6 due to the fact that it has lesser impact on prolactin and Cortisol; at least as compared to GHRP 2.
