Now these artificially manufactured compounds can replicate certain hormones within the human body to signal or trigger certain effects. Whereas GH (a total protein hormone of 191 amino acids), can attach to receptors at multiple sites within the body to influence different effects (say muscle cells, bone cells and fats cells, to name but three), shorter peptides have been isolated to trigger certain effects in a specific area/s. Imagine it’s like having a full tool box which you can mend a whole car with, but then you take certain tools out for different jobs and keep them apart in separate drawers which do specific jobs…sort of.
Perhaps, the primary reason why peptides are used in bodybuilding has to do with how they improve growth hormone production. It is no longer a secret how HGH helps to improve lean muscle mass. Your body uses peptides to promote increased secretion of HGH. They, therefore, make it easier for interested persons to achieve bigger muscle mass. Their usage may help enhance strength. It is even said that these small amino acid chains could boost testosterone production.
The most important initial historical time points in the development of the enlarging ghrelin system were 1973, 1976, 1982, 1984, 1990, 1996, 1998, and 1999 during which the following sequentially occurred: isolation of somatostatin; discovery of unnatural growth-hormone-releasing peptides (GHRPs); isolation of growth-hormone-releasing hormone (GHRH); hypothesis of a new natural GHRP different from GHRH; GHRP+GHRH synergism in humans; discovery of the growth hormone secretagogue GHS/GHRP receptor; cloning of the receptor; isolation; and identification of the new natural endogenous GHRP ghrelin.1
In 1984, a synthetic hexapeptide, His-d-Trp-Ala-Trp-d-Phe-Lys-NH2 (GHRP-6), was identified by Bowers and colleagues. This hexapeptide was shown to potently stimulate GH release in vitro and in vivo by an unknown mechanism. Because of its poor oral bioavailability (0.3%) and short half-life (20 min) in human serum, GHRP-6 was selected only as a structural model to design a nonpeptide mimetic. Based on the structure–activity relationships (SARs) of GHRP-6, the nonpeptidyl growth hormone secretagogue (GHS) L-692,429 was identified by Smith et al. in 1993. This nonpeptidyl GHS synergizes with GHRP-6 to stimulate GH release and cAMP production, accompanied by a significant increase in intracellular calcium concentration ([Ca2 +]i), indicating that this nonpeptidyl GHS acts through a distinct signal transduction pathway. In 1995, a potent oral GHS L-163,191 (MK-0677) was reported by Patchett et al. This agent was found to have excellent oral bioavailability and specificity in its release of GH, without significant effect on plasma levels of other hormones such as aldosterone, luteinizing hormone, thyroxine, and prolactin.
Broadly speaking, it’s long been a widespread view that fasting can in many instances provide healthful effects beyond simple fat loss. It’s speculative to say that increased ghrelin levels must be a major cause of such effect (if granting the effect), but it’s entirely consistent with the scientific literature that such elevation of ghrelin levels may have health benefits. Appropriate-dosed and cycled GHRP use may at least partially provide such benefits, particularly with regard to anti-inflammatory and healing effect.

SARMs can be taken orally as well, and regularly stacked together for improved results. They have been around for years now (although only recently gaining popularity) and there is a wealth of studies done on them as well. Because some GHRP’s are equally effective as others in increasing GH but differ in effect on hunger or ACTH stimulation, it seems likely that there may be differences in ghrelin receptors between different tissues, or differences in function (for example with cofactors.) This is the most likely explanation for GHRP-6 being effective in stimulating hunger and helping heal tendinitis, while GHRP-2 stimulates hunger less and may have less value for healing.
The use of peptides is not necessarily a case of one or the other. While you can derive benefit from using either GHRH or GHRP, the effects can be amplified by using both. This, for one, will help to better keep levels of somatostatin under control. The amount of that hormone is an issue you will be forced to contend with when using only GHRH. This is because it is known to inhibit growth hormone.
This peptide is a modified fragment of hGH which contains the portion of the molecule that is believed to be responsible for hGH’s anti-obesity effects. The peptide has been shown to increase fat burning without the increase in blood sugar and growth rate that has been seen with hGH itself. AOD 9604 has been deemed safe for chronic use by the FDA, receiving Human GRAS status in 2014. The best way to summarise how well SARMs (and peptides for that matter) work is to point out that they have been banned by WADA (the world anti-doping authority) for competitive sports because they give an unfair advantage to athletes. Luckily they are legal to purchase so if Sarms can do that for athletes imagine how much they can help ordinary folk.

GHRP-6 brings about the effects you’d expect from heightened Growth Hormone and IGF-1 levels; increased fat loss and muscle building. It’s worth remembering that Growth Hormone and IGF-1 will not only promote greater muscle hypertrophy (enlargement of existing muscle fibres) but will also cause muscle hyperplasia – an increase in the actual number of muscle cells.
Paracetamol has long been considered very safe, without the risks of gastric injury associated with aspirin and NSAIDs. But there are distinct risks of liver injury, usually following overdose situations. In response many international regulatory authorities have taken steps to reduce the pack sizes of paracetamol, and to restrict release in some environments to pharmacies. In the USA, FDA has required prescription acetaminophen, when it is usually combined with an opioid, to reduce the dose per dose unit to 325 mg, but without reducing the maximal daily dose. No change of dosing in the USA has yet come for OTC acetaminophen. Use of paracetamol should be kept to a minimum in patients with underlying liver and renal disease. It can reduce the effects of lithium, ACE inhibitors, beta blockers and methotrexate. However, it remains one of the safest and most effective analgesic drugs, particularly in the elderly where the risks of gastric bleeding with NSAIDs are more common, and carries minimal side effects.
GH-releasing peptides (GHRPs) are synthetic peptides that like GHRH act directly on pituitary somatotrophs to stimulate GH release. Growth hormone (GH) release is stimulated by a variety of synthetic secretagogues, of which growth hormone-releasing hexapeptide (GHRP-6) has been most thoroughly studied; it is thought to have actions at both pituitary and hypothalamic site.
There are some alternatives to GHPR-6 in the market which are easier to procure and considered legal in many countries. Natural supplements consisting of amino acids which are safer and non-hormonal when brought together may facilitate the increase of natural manufacturing of HGH. The effectiveness of these alternatives are likely to be inferior to the real deal but you should consider them if you are unlikely to get your hands on that precious prescription. Do a bit of research on your own and try finding the best option for you.
The evidence derived from these experiments supports the notion that CD36 is an active and approachable receptor to modulate the healing process. Here we have observed that CD36 occupation by GHRP-6 attenuates wound inflammation, accelerates wound closure, and above all improved wound’s esthetic outcome by impacting ECM proteins accumulation. To our knowledge these findings are unprecedented for GHRP-6 within the context of cutaneous healing.
Following the preliminary histological data, suggesting a reduction of wound inflammation and a far more organized ECM, we addressed the gene expression study toward inflammatory and profibrogenic markers. We primarily examined Cd36 expression following topical GHRP-6 application and found that peptide reduced its receptor expression () (Figure 4). Furthermore, the treatment significantly reduced Adam17 expression () and approached to significantly reduce Tnf (), which may partially contribute to explaining the substantial reduction of infiltrated inflammatory cells within the wound bed (Figure 4).

A multicenter study comparing the oral GH secretagogue macimorelin with arginine/GHRH found it to be safe, convenient, and of comparable efficacy (82% sensitivity, 92% specificity, and 87% accuracy in diagnosing adult GHD), with a GH cut-off point of 6.8 μg/L for patients with a body mass index (BMI) <30 kg/m2 and 2.7 µg/L for patients with a BMI >30 kg/m2 [268].

So… now we get down to the individual peptides themselves. I will not go into length with a profile for each one, but instead I will provide two charts that should help in understanding which peptides are GHRH’s and which ones are GHRP’s. Of course, IGF and MGF have their own respective spots but do not have the synergy when combined like GHRP and GHRH.

Hexarelin is a peptide that is derived from GHRP 6, but has been optimized to enhance its metabolic stability. Like the other GHSs, hexarelin increases hGH production, resulting in increased muscle mass, bone density, skin elasticity, and decreased body fat. Unlike the other GHRPs, however, hexarelin does not lead to a substantial increase in ghrelin and therefore does not cause the same appetite stimulation. This peptide has been further promoted for its cardioprotective and regenerative action as well. Hexarelin would be an ideal choice for those looking to benefit from increased growth hormone without appetite stimulation.

Similar to other enhancers, it is observed that administrating GHRP-6 along with insulin gets an increased GH response. However, in presence of elevated glucose levels, GHRP-6 does not work well. As a result, consuming carbohydrates or dietary fats before administrating GHRP-6 is a bad idea. Thus the dose should be taken two hours after your last meal and at least thirty minutes before your next meal. Also, GHRP-6 has saturation points. Well, personally I used 150mcg injected directly into the joints or areas that I've had any niggling injuries, the localized effect it has on collagen growth is nothing short of astounding. After using GHRP-6, I have personally recovered from a full pectoral tendon tear, where the tendon ripped right of the humerus bone; in fact, it's now in better shape than it was prior to the injury. Despite the controversies, some scientists continued with additional studies and again proved IGF-1 to actually prolong life…at least in worms.  Then, in 2001, scientists discovered that the use of IGF-1 resulted in a proliferation of cancer cells, especially throughout the breast and colon, and a 2012 study found that both too much or too little IGF-1 could contribute to dying from cancer; implying that IGF-1 actually helped patients with terminal cancer live longer.
Despite their potent and reproducible GH-releasing activity, the clinical use of GHRPs as orally active growth-promoting agents and anabolic antiaging drugs remains to be confirmed.13 Accordingly, the early years’ enthusiasm as an alternative for GH replacement therapy faded away soon after their discovery.16 Nevertheless, it is likely that the myocardial, vascular, and multiorgan expression of the GHRP receptors may have contributed to reinforce the cardiovascular application stream of these peptides.
