TO-2 hamster model of DCM was characterized by progressive LV dilation, LV wall thinning, LV systolic dysfunction, and reduced life span; both GHRP-2 and GHRP-6 ameliorated all the dysfunctional ventricular parameters and reduced the progression of the DCM.34 We also examined the potential impact of GHRP-6 in a rat model of DCM/heart failure induced by doxorubicin (DX). The concurrent administration of GHRP-6 was undertaken with the purpose to study the potential prophylactic impact before the cardiac function demise. As part of the prolonged treatment with DX, the concurrent administration of GHRP-6 completely prevented failure of cardiac function, which was evaluated as the percentage of ejection fraction by echocardiography (Figure 2, prevention). This effect significantly increased the survival of animals. Similar results were obtained in the therapeutic administration schedule, with functional recovery of cardiac muscle to physiological levels (Figure 2, regression), also attenuating systemic damages and, consequently, decreasing the mortality rates of rats. In the experimental model of DX-induced cardiac and systemic damage, GHRP-6 additionally attenuated various extracardiac damages observed in the renal tubular and bronchoalveolar epithelial structures as in the hepatic parenchyma.35
I started taking Ipamorelin and CJC 1295 without DAC yesterday. I dont see many logs and i see a lot of people wondering what kind of results you can get wiith these peptides. Ghrelin has been linked to inducing appetite and feeding behaviors. Circulating ghrelin levels are the highest right before a meal and the lowest right after. Injections of ghrelin in both humans and rats have been shown to increase food intake in a dose dependent manner. So the more ghrelin that is injected the more food that is consumed. However, ghrelin does not increase meal size, only meal number. Ghrelin injections also increase an animals’ motivation to seek out food, behaviors including increased sniffing, foraging for food, and hoarding food. The potent biologic effects of GHRPs and the identification of the GHS-R suggested the existence of a natural ligand for the receptor that is involved in the physiologic regulation of GH secretion. The acylated peptide ghrelin, produced and secreted into the circulation from the stomach, is this ligand (Fig. 7-22). The effects of ghrelin on GH secretion in humans are identical to or more potent than those of the non-natural GHRPs (see Fig. 7-20). In addition, ghrelin acutely increases circulating PRL, ACTH, cortisol, and aldosterone levels. There is debate concerning the extent and localization of ghrelin expression in the brain that must be resolved before the implications of gastric-derived ghrelin in the regulation of pituitary hormone secretion are fully understood. Furthermore, post-translational processing of pro-ghrelin gives rise to a second neuropeptide, obestatin, which may also have functional roles in activity of the GH/IGF-1 axis and metabolism. A proposed role for pro-ghrelin peptides in appetite and the regulation of food intake is discussed in Chapter 35.
These peptides are inhibitors of a protein called myostatin. Myostatin is secreted by muscle cells and acts to essentially block the development of new muscle fibers and, thus, the development of lean muscle mass. In fact, individuals who have mutations in the gene coding for this protein have significantly more muscle mass and enhanced strength. The administration of follastatin peptides can generate enhanced muscle mass and strength, and has been found especially useful in patients suffering from muscle-wasting diseases or who have difficulty gaining muscle.
GHRP-6 is a peptide from the category of growth hormone releasing peptides, ie growth hormone-releasing peptides. The most frequent use of these peptides is an extreme increase in the body's growth hormone production. The main application area of GHRP-6 is an increase in growth hormone levels, which also results in an increase in IGF1 levels. This also radically supports fat loss as well as muscle building. In general, GHRPs are used as an alternative to growth hormones or even combined with growth hormones to achieve virtually a double effect, synthetic and natural. Reasons for favoring GHRP-6 versus other GHRPs are its appetite-stimulating effect, although present, but weaker than GHRP-2 and its ability to reduce inflammation and assist healing of injuries such as tendonitis. Serum ghrelin levels vary as a function of energy balance. Ghrelin levels are increased in anorexia and decreased in obesity.78 Thus, it is possible that ghrelin may be an important player in food intake behavior and perhaps in chronic over- and under-nutrition as well.9 Because of its dual effects, ghrelin may be a critical hormonal signal of nutritional status to the somatotropic axis, playing a role in integrating energy balance with the growth process.10

In studies of the opioid control of GH secretion, several peptide analogues of met-enkephalin were found to be potent GH secretagogues. These include the GH-releasing peptide GHRP-6 (Fig. 7-21), hexarelin (His-d2MeTrp-Ala-Trp-dPhe-Lys-NH2), and other more potent analogues including cyclic peptides and modified pentapeptides. Subsequently, a series of nonpeptidyl GHRP mimetics were synthesized with greater oral bioavailability, including the spiropiperidine MK-0677 and the shorter acting benzylpiperidine L-163,540 (see Fig. 7-21). Common to all these compounds, and the basis of their differentiation from GHRH analogues in pharmacologic activity screens, is their activation of phospholipase C and inositol 1,4,5-trisphosphate. This property was exploited in a cloning strategy that led to the identification of a novel GPCR GHS-R that is highly selective for the GH secretagogue class of ligands. The GHS-R is unrelated to the GHRH receptor and is highly expressed in the anterior pituitary gland and multiple brain areas, including the medial basal hypothalamus, the hippocampus, and the mesencephalic nuclei that are centers of dopamine and serotonin production.
The known side effects of IGF-1 injections include jaw pain, facial and hand swelling and heart-rhythm disturbances, especially if doses of more than 100 micrograms (mcg) are injected. Exceeding 100mcg of IGF-1 can actually cause your heart to stop beating and blood pressure to drop dramatically. This is caused by an IGF-1-induced drop in blood phosphate levels, and in the bodybuilding community is often prevented by administering phosphate with the IGF-1.
A remarkable specific (125)I-Tyr-Ala-hexarelin binding was observed in the human cardiovascular system where the highest binding levels were detected in ventricles, followed by atria, aorta, coronaries, carotid, endocardium, and vena cava. In other experiments on H9c2, cardiomyocyte-specific GHRP binding was found along with a potent antiapoptotic activity.3 The primarily investigated receptor was the growth hormone secretagogue receptor type 1a (GHS-R1a), which was detected in isolated human cardiomyocytes, myocardium, and aorta samples.17 It has been recently shown that GHS-R1a is a sort of “promiscuous receptor” involved in many systems and behavioral patterns such as reward, feeding, and memory, which makes it an attractive pharmacological target.18 Years later, the synthetic GHRP hexarelin was acknowledged as a ligand of another protein identified as CD36, a scavenger receptor that is expressed in various tissues, including monocytes/macrophages and the endothelial microvasculature. Activation of CD36 in perfused hearts by hexarelin was shown to increase coronary perfusion pressure in a dose-dependent manner. Contrariwise, this effect was lacking in hearts from CD36-null mice and hearts from spontaneous hypertensive rats genetically deficient in CD36.19,20 Thus, it is currently accepted that two cardiac receptor subtypes mediate the pharmacological actions of GHRP-6, GHRP-2, and hexarelin.21,22
Myocardial ischemia/reperfusion damage entails multiple molecular and biochemical mechanisms that each alone is sufficiently injurious to disturb an organ whose mechanical performance is dependent upon the stability of ionic/electrical pumps. Oxidative stress, intracellular calcium overload, pH changes, mitochondrial dysfunction, inflammation, and excessive neurohormones are part of an interactive and self-perpetuating continuum of the myocardial injury cascade (Figure 1). The evidences obtained along the years of experimental screening of the synthetic GHRP suggest that each single member of this family of peptides is able to simultaneously counteract different injurious operators in the myocardial ischemic event.

It should be noted right off the bat that GHRP-6 doses are often normally (and ideally) combined with doses of a GHRH analogue, such as Mod GRF 1-29 (CJC-1295 without DAC) due to the synergistic effects and compatibility between the two, as previously mentioned in this profile. With that being said, the proper GHRP-6 doses do not change whether or not it is utilized with a GHRH. If an effective GHRP-6 dose is 100mcg, for example, then 100mcg of GHRP-6 should be administered whether the user is utilizing it alone or with Mod GRF 1-29. The term/phrase “saturation dose” or “saturation doses” can be heard a lot when peptides are discussed. A saturation dose is defined as a dose that will completely (or near completely) saturate the peptide’s target receptors. In GHRP-6’s case, this means the Ghrelin receptors located on the hypothalamus and the anterior pituitary.

by Bill Roberts – GHRP-6 is an injectable peptide in the category of growth hormone releasing peptides, or GHRP’s. The most common use of these peptides is to increase GH production. Other peptides in this category include GHRP-2, hexarelin, and ipamorelin. With regard to increasing GH, all of these work similarly, and there is no need or advantage to combining them. Instead, the one most suited for the particular case is chosen.
Similar to other enhancers, it is observed that administrating GHRP-6 along with insulin gets an increased GH response. However, in presence of elevated glucose levels, GHRP-6 does not work well. As a result, consuming carbohydrates or dietary fats before administrating GHRP-6 is a bad idea. Thus the dose should be taken two hours after your last meal and at least thirty minutes before your next meal. Also, GHRP-6 has saturation points. As a result, you want to put a healthy interval between two doses so that your receptors are clear. The best schedule is one dose upon waking up, one post workout, and one before sleeping.

The effect of GHSs on GH release is dose dependent and more reproducible than that of GHRH. The peptide GHSs (e.g., GHRP-6, GHRP-1, GHRP-2, and hexarelin) and the nonpeptide GHSs differ in terms of their pharmacokinetics. The nonpeptides MK-0677 and macimorelin have been developed specifically as orally active agents. The peptidyl GHSs are also active PO, but only at doses several hundred times higher than that required when administered IV.
A seminal report by a Merck Research Laboratories group dated 2003 demonstrated for the first time that chronic treatment with GHRP-6 (21 days) prevented sudden death in a canine model of DCM and subsequently subjected to acute myocardial infarction (AMI). In the meantime, the mortality rates for the vehicle and GH-treated groups were about 50%. Although the authors do not precise the mechanism underlying the 100% survival in the GHRP-6 group, an enhanced regional myocardial compensatory function of the nonischemic zone was assumed.40 This notion could be validated at least in part by the fact that the cardiotropic effects shown by GHRP-1, GHRP-2, GHRP-6, and hexarelin in cardiomyocytes and isolated, denervated, perfused hearts are mediated by an elevation of Ca2+ influx through the voltage-gated calcium channel, triggering Ca2+ release from thapsigargin-sensitive intracellular stores, which translated in a positive inotropic response without a chronotropic effect.41 More recent data confirm the ability of hexarelin and other secretagogue peptides that bind and activate the GHS-R1a, to control the cardiac action potential and reduce apoptosis of cardiomyocytes, derived from isolated hearts subjected to ischemia/reperfusion episodes.42

As is the case with ghrelin, GHRP 2 has the ability to stimulate secretion of other growth hormones and increase food consumption. When you start taking in GHRP 2 at regular intervals the level of growth hormones being released in your body increases considerably. Another interesting attribute with GHRP 2-based supplements is that they are anti-inflammatory. But the extent of this feature will vary from one person to another as the pituitary somatotrophs of an individual will respond differently to different receptor and so on.
