SARMs stimulate androgen receptors specifically in muscle and bone cells, hence assisting with muscle and bone growth, while having little effect on the other cells in the body (unlike regular steroids). It is both impractical and impossible to categorize GHRP-6 doses into the typical three tiers of users (beginner, intermediate, and advanced) due to the inherent nature of the type of substance and hormone. The saturation doses of GHRP-6 should elicit significant increases in HGH levels in the body in a pulsatile manner that is adequate enough to ensure any performance and physique enhancing goals desired by any individual. The difference in which goals might be more attainable than others is the frequency of dosing. Despite their potent and reproducible GH-releasing activity, the clinical use of GHRPs as orally active growth-promoting agents and anabolic antiaging drugs remains to be confirmed.13 Accordingly, the early years’ enthusiasm as an alternative for GH replacement therapy faded away soon after their discovery.16 Nevertheless, it is likely that the myocardial, vascular, and multiorgan expression of the GHRP receptors may have contributed to reinforce the cardiovascular application stream of these peptides.
The effect of GHSs on GH release is dose dependent and more reproducible than that of GHRH. The peptide GHSs (e.g., GHRP-6, GHRP-1, GHRP-2, and hexarelin) and the nonpeptide GHSs differ in terms of their pharmacokinetics. The nonpeptides MK-0677 and macimorelin have been developed specifically as orally active agents. The peptidyl GHSs are also active PO, but only at doses several hundred times higher than that required when administered IV.

GHRP-6 is a peptide from the category of growth hormone releasing peptides, ie growth hormone-releasing peptides. The most frequent use of these peptides is an extreme increase in the body's growth hormone production. The main application area of GHRP-6 is an increase in growth hormone levels, which also results in an increase in IGF1 levels. This also radically supports fat loss as well as muscle building. In general, GHRPs are used as an alternative to growth hormones or even combined with growth hormones to achieve virtually a double effect, synthetic and natural. Our first human GHRP-6 studies in normal young men were performed in collaboration with Michael Thorner (Bowers et al., 1990). These studies (Fig. 1.7, left panel) revealed that iv bolus GHRP-6 released GH and, when given together with GHRH, released GH synergistically. One of the most characteristic and consistent in vivo actions of GHRPs in various animals as well as humans of both sexes and all ages is the synergistic release of GH when GHRP is administered concomitantly with GHRH by iv bolus. Subsequently, this was also found for continuous 24/7 subcutaneous (sc) infusion. Also recorded in Fig. 1.7, right panel, is the comparative GH-releasing effects of iv bolus GHRP-6, -1, -2, and GHRH in normal young men. The potency of the three GHRPs we developed over several years was increasingly effective in releasing GH, and each released more GH than GHRH in normal young men. In addition, this was also found to occur in normal young women (Bowers, 1996).
Thymosin beta 4 (Tβ4) is the predominant form of thymosin in our bodies. It has been found in high concentrations in wound tissue and certain blood cells involved in clotting, signifying its important role in the healing process. In fact, recent studies have revealed that the first gene to be upregulated after an injury is the Tβ4 gene. As the body begins the recovery process, Tβ4 aids in the creation of new vessels in the injured area, which carry blood, nutrients, and reparative substances to the site. Tβ4 also has anti-inflammatory properties, and works to decrease the amount of inflammatory substances, called cytokines. Inflammation plays a large role in many of the symptoms associated with a large number of conditions (i.e., Lyme disease, CFS, FM, autoimmune diseases, infections, etc.), making the potential impact of Tβ4 quite extensive.
The scheduling of paracetamol and caffeine when combined in a compound analgesic as the only active ingredients was again reviewed by the NDPSC at its 57th Meeting in October 2009 after the Committee had received a request to reconsider the scheduling on the grounds of potential toxicity if used in excess. Lactating nipples: GHRP-6 side effects include that of increased Prolactin secretion, which can result in lactation from the nipples. This is, however, a rare occurrence but it can happen in sensitive individuals. GHRP-6 exhibits the ability to induce secretion of Cortisol and Prolactin[3], but studies have shown that the Prolactin and Cortisol increases in most test subjects were not altered at all at GHRP-6 doses of 100mcg or less[4] [5]. Doses above 100mcg are said to increase Prolactin secretion, though minimally, and at these minimal levels lactation should not present itself. However, some users have reported lactation which can be the result of a sensitive individual or the result of much higher doses of GHRP-6. Prolactin can be lowered through the use of a Prolactin antagonist such as Cabergoline, Pramipexole, Bromocriptine, and even vitamin B6.

Three of the submissions did not support the proposal highlighting the impact the change in scheduling would have on product currently on the market, industry, pharmacists and consumers. Two submissions noted that there has not been a history of concern with this combination of substances. One submission, referring to the NEJM article, believed that a lack of information about the study means that it cannot be relied upon as there is not a meaningful assessment of the results.

RT-PCR experiments shed light on the molecular mechanisms by which GHRP-6 appeared to modulate the fibrotic response. Among the genes studied (Table 1), GHRP-6 proved to significantly reduce TGFB1 and CTGF () expression, with no effect on PDGFB gene expression. An unexpected finding was that MMP3 appeared significantly reduced in the GHRP-6-treated wounds (). Most meaningfully is that PPARG expression became significantly elevated with GHRP-6 treatment (), as compared to placebo-treated wounds (Figure 7).
Similar to other enhancers, it is observed that administrating GHRP-6 along with insulin gets an increased GH response. However, in presence of elevated glucose levels, GHRP-6 does not work well. As a result, consuming carbohydrates or dietary fats before administrating GHRP-6 is a bad idea. Thus the dose should be taken two hours after your last meal and at least thirty minutes before your next meal. Also, GHRP-6 has saturation points. As a result, you want to put a healthy interval between two doses so that your receptors are clear. The best schedule is one dose upon waking up, one post workout, and one before sleeping.
Growth hormone releasing peptide (GHRP) 2 is a type of peptide therapeutic that mimics the effects of ghrelin, the “hunger hormone”. Ghrelin is a hormone that helps regulate appetite as well as energy distribution and rate of use, or metabolism. In the 1980’s, ghrelin was discovered to be the body’s natural ligand (or binding molecule) of the GHRP receptor in the anterior pituitary. This was a significant discovery, as it highlighted the role of ghrelin in hGH secretion and growth regulation. Modern biotechnology has used this knowledge to develop peptides that can be administered to mimic ghrelin’s hGH stimulation, but in a more targeted fashion. GHRP 2 is one such peptide, stimulating hGH secretion by 7-15 times, increasing appetite and meal initiation, while also decreasing fat mass and cholesterol. GHRP 2 is ideal for patients who are hypercatabolic, due to critical illness, cancer, AIDS, etc. It should be noted that GHRP 2 can also increase levels of prolactin, aldosterone, and cortisol.

also using a dose of 400mcg for the IPAM is really a waste of your peptides as anything above saturation dose will give diminished returns, saturation dose(1mcg per kg) is a dose that will give maximum return, if you double the saturation dose then you will not get double the GH pulse in fact no where near that, then the more you use the less added benefit you will get.
IGF-1 also increases the activity of muscle protein synthesis and the activity of muscle stem cells (also called satellite cells) for repair of damaged muscle. This is probably why intense weight training is one primary stimulus for a natural release of IGF-1 in muscle. As a matter of fact, exercise researchers have found that systemic IGF-1 normally produced in the liver isn’t even required for this type of muscle repair, as other IGF-1 forms produced by your own muscles during and post-exercise allows for adequate muscle tissue repair.
The conclusion comes down to which peptides should you use. This is going to depend on personal use and experience. Whether you are into bodybuilding, gaining an edge in sports or just looking for health and wellness and anti aging properties, peptides offer a little something for everyone. There are many peptides that are not covered below, but as an introduction into the peptide world these are the most generic and widely used. As with any supplement or drug, please do your own research before diving into using peptides.
Of particular note is the variable chemistry of GHRPs, which consist of three major chemical classes including peptides, partial peptides, and nonpeptides, all of which appear to act via the same receptor and cellular mechanisms. Generally, most GHRPs are active by all routes of administration, specifically intravenously (IV), subcutaneously (SC), orally, intranasally, and intracerebroventricularly (IVC), which supports their possible broad future clinical utility. From evolutionary studies starting with the zebrafish, the natural receptor and hormone have been present for hundreds of years, underscoring the fundamental evolutionary and functional importance of the ghrelin system. GHRPs were well established to act directly on both the hypothalamus and pituitary several years before the GHS receptor assay.23

These amino acid chains communicate with the body to produce or release growth hormone. Hence a secretagogue causes the body’s own natural processes to produce growth hormone. The subcutaneous injection route of growth hormone stimulation is a preferred route to help slow down age and environmental reductions in growth hormone levels.
Administration of peptides is normally subcutaneous or intramuscular. Peptides come as a fine white and delicate powder that must be reconstituted with bacteriostatic water or medical grade saline. An insulin syringe should always be used to administer the dose. When targeting muscles, look for a place where the layer of skin and fat are lean. Mixing two peptides in the same syringe is totally fine but I personally would advise not drawing/mixing doses and storing pins for future use.
Immunohistochemical determination of CD31 expression (platelet endothelial cell adhesion molecule-1, PECAM-1) was conducted as this is a marker protein of mature vascular endothelium [19]. Sections (5 μm) were mounted on chromalum-coated slides, dewaxed, rehydrated, rinsed, and washed in PBS 1x solution for 30 min. Once endogenous peroxidase was quenched, the specimens were treated with target retrieval solution (Dako) equilibrated at 99°C. Tissue samples were then incubated for 40 min with 1/50 dilution of anti-CD31 antibody (Abcam 28364, USA) in background reducing solution (Dako). The immunohistochemical reactions were carried out using the labelled streptavidin/biotin-horseradish peroxidase conjugate method, according to the manufacturer’s instructions (Dako). The peroxidase reaction was developed with diaminobenzidine and counterstained with hematoxylin.
In more recent years, these data were further substantiated using again the TO-2 hamster DCM biomodel in which GHRP-2 reduced the progression of LV remodeling, dysfunction, and the ensued myocardial fibrosis by an antioxidant mechanism.36 The abovementioned myocardial fibrotic process amelioration reveals an additional potential use for GHRP in an unmet medical need. Chronic treatment with hexarelin in spontaneously hypertensive rats, in addition to decreasing ventricular hypertrophy, diastolic dysfunction, and high blood pressure, significantly reduced cardiac fibrosis by decreasing interstitial and perivascular myocardial collagen deposition and myocardial hydroxyproline content. Mechanistically, hexarelin treatment increased matrix metalloproteinase (MMP)-2 and MMP-9 activities and decreased myocardial mRNA expression of tissue inhibitor of metalloproteinase (TIMP)-1.37
Though there are a lot of similarities between the two peptides, there is no denying the fact that they also have vast differences. Besides having differences in the release of growth hormones and stimulation of pituitary glands, GHRP 2 is known to have better control in the release of prolactin and Cortisol. That’s not all; GHRP 2-based supplements are relatively cheaper as compared to other supplements that are available in the market, including the ones that are GHRP-6 based. There are a few individuals that opt for GHRP 6 due to the fact that it has lesser impact on prolactin and Cortisol; at least as compared to GHRP 2.
