GHRP-6 brings about the effects you’d expect from heightened Growth Hormone and IGF-1 levels; increased fat loss and muscle building. But gene-therapy experiments have also resulted in patient deaths. The use of such therapies can cause the human body to experience fatal immune reactions to the vectors used to place the gene in the body. Another danger of gene therapy is an inability to control the expression of the gene, which could translate into a rapidly spreading cancer. Or the expression of the gene could spread from skeletal muscle into heart muscle, resulting in excessive heart muscle growth (known as left ventricular hypertrophy, or “athlete’s heart) that can cause premature heart failure.
Total RNA was purified according to TRI Reagent standard procedure (Sigma, USA), following digestion with RQ1 DNase I (Promega, USA) to remove contaminating genomic DNA. Afterward, 500 ng of DNA-free RNA was reverse transcribed using Omniscript RT kit (Qiagen, Germany) with oligo-dT primer. The RT reaction was performed at 42°C for 60 min. PCR mixtures contained 1 μL cDNA, 1 μL of each primer (10 μM), and 12.5 μL 2x Taq MasterMix (Qiagen, Germany) in a final volume of 25 μL. Specific sense and antisense primers, annealing temperatures, and number of repeating cycles for both studies are referred to in Table 1. Amplifying conditions were performed as follows: a first step of 95°C for 5 minutes, thereafter repeating cycles comprised of 95°C for 30 seconds, specific annealing temperature for 30 seconds and 72°C for 30 seconds, and a final extension step of 5 minutes at 72°C. PCR bands (8 μL of PCR product plus 2 μL of gel loading buffer) were resolved on 1.5% (w/v) agarose gel electrophoresis and visualized under ultraviolet light subsequent to being stained with ethidium bromide. PCR products were quantified using the Kodak ID 3.6 software package (Kodak Inc, USA). Beta-2 microglobulin was used as housekeeping gene for normalization.

These peptides are inhibitors of a protein called myostatin. Myostatin is secreted by muscle cells and acts to essentially block the development of new muscle fibers and, thus, the development of lean muscle mass. Ghrelin levels are increased in anorexia and decreased in obesity.78 Thus, it is possible that ghrelin may be an important player in food intake behavior and perhaps in chronic over- and under-nutrition as well.9 Because of its dual effects, ghrelin may be a critical hormonal signal of nutritional status to the somatotropic axis, playing a role in integrating energy balance with the growth process.10

The potent biologic effects of GHRPs and the identification of the GHS-R suggested the existence of a natural ligand for the receptor that is involved in the physiologic regulation of GH secretion. The acylated peptide ghrelin, produced and secreted into the circulation from the stomach, is this ligand (Fig. 7-22). The effects of ghrelin on GH secretion in humans are identical to or more potent than those of the non-natural GHRPs (see Fig. 7-20). In addition, ghrelin acutely increases circulating PRL, ACTH, cortisol, and aldosterone levels. There is debate con­cerning the extent and localization of ghrelin expression in the brain that must be resolved before the implications of gastric-derived ghrelin in the regulation of pituitary hormone secretion are fully understood. Furthermore, post-translational processing of pro-ghrelin gives rise to a second neuropeptide, obestatin, which may also have functional roles in activity of the GH/IGF-1 axis and metabolism. A proposed role for pro-ghrelin peptides in appetite and the regulation of food intake is discussed in Chapter 35.
For example, insufficient protein or calories can cause IGF-1 to plummet, while ample calories can cause IGF-1 to increase. For example, one study of women who fed with excess calories over and above their normal metabolic rate noted a 19% increase in IGF-1 after two weeks of overfeeding, with 46% of the weight gain from  lean mass and 54% from bodyfat. Fasting insulin doubled in these women, and testosterone levels also significantly increased.

Ipamorelin is a pentapeptide, meaning that it is composed of five amino acids, that mimics the body’s natural GH release.  Ipamorelin is a growth hormone releasing peptide (GHRP) and analogue of the hormone Ghrelin. It induces GH release and increases the number of somatarophs(cells responsible for GH release) in a GH pulse by suppressing somatostatin.

In a study designed to assess the effect of both the estrogen and GHRP-6 on the cardiovascular and metabolic diseases in ovariectomized (OVX) rats, Elbassuoni, et al found that although GHRP-6 failed to produce significant change in body weight gain and food intake, it clearly reversed the effect of OVX on fasting serum glucose, insulin, insulin resistance, and the assessed lipid fractions. They concluded that the effect of GHRP-6 on improving dyslipidemia after OVX was even more potent than that of estrogen.12 Furthermore, the mechanism of action of GHRP-6 has been more extensively studied in experimental models with obese subjects, and was shown to be a powerful GH releaser in obesity, and to release GH independently of the hypothalamic factors (GHRH and somatostatin).13

Specifically, T α 1 has been shown to enhance the function of certain immune cells called T and dendritic cells. This is very important to anyone with a depressed immune system or suffering from an infection, as these white blood cells play pivotal roles in the body’s defense process. T cells, for example, come in two forms: killer and helper T cells. Killer T cells are responsible for hunting down and destroying our body’s own cells that are cancerous or infected with bacteria or viruses. GHRP-6 side effects that are the result of the HGH increases include: flu-like symptoms, joint pain, and carpal tunnel syndrome, headaches, and bloating and water retention. I stopped the colostrum and my ” symptoms ” subsided, seems I have a moderately enlarged prostate which doesn’t run in my family on either side, my question is could the colostrum possibly cause the prostate to enlarge due to the igf-1 at a certain age,? due to a possible decline in testosterone, or could the benefits of colostrum outweigh the prostate issue?
High testosterone at this stage will accelerate the process. The SARMs are not testosterone, and don’t get metabolised into DHT (nor estradiol). The SARMs selectively bind to the androgen receptor in muscle and bone and amplify the effect of testosterone and DHT there, while not amplifying the effect on other tissue ie skin, prostate. However, through inheritance, if you have hair androgen receptors that are similar to muscle/bone androgen receptors, then SARMs can amplify the androgen message in the hair follicles, and if the inherited androgen sensitivity is activated, it could lead to accelerated male pattern baldness. This is a very rare variation, and while possible, is uncommon. There are no tests available to determine SARMs effect on your hair follicles, nor to determine when your genetic androgen sensitivity in hair follicles will activate.
A remarkable specific (125)I-Tyr-Ala-hexarelin binding was observed in the human cardiovascular system where the highest binding levels were detected in ventricles, followed by atria, aorta, coronaries, carotid, endocardium, and vena cava. In other experiments on H9c2, cardiomyocyte-specific GHRP binding was found along with a potent antiapoptotic activity.3 The primarily investigated receptor was the growth hormone secretagogue receptor type 1a (GHS-R1a), which was detected in isolated human cardiomyocytes, myocardium, and aorta samples.17 It has been recently shown that GHS-R1a is a sort of “promiscuous receptor” involved in many systems and behavioral patterns such as reward, feeding, and memory, which makes it an attractive pharmacological target.18 Years later, the synthetic GHRP hexarelin was acknowledged as a ligand of another protein identified as CD36, a scavenger receptor that is expressed in various tissues, including monocytes/macrophages and the endothelial microvasculature. Activation of CD36 in perfused hearts by hexarelin was shown to increase coronary perfusion pressure in a dose-dependent manner. Contrariwise, this effect was lacking in hearts from CD36-null mice and hearts from spontaneous hypertensive rats genetically deficient in CD36.19,20 Thus, it is currently accepted that two cardiac receptor subtypes mediate the pharmacological actions of GHRP-6, GHRP-2, and hexarelin.21,22
