I have questions about combined therapy of CJC 1295 and Ipamorelin at the same time on a daily basis for both. The compounding pharmacies do not clearly state whether the CJC is with or without DAC. If it is the CJC with DAC, which sustains elevated GH and IGF-1 for several days, would taking it nightly in conjunction with the Ipramorelin, that is suggested to be taken TID but is being recommended only once at night, be over-stimulatory? If the CJC is without DAC, why take two pepetides simultaneously ,that have similar effects? I am just not clear why taking a daily dose of CJC with Ipamorelin as a single dose is better than taking the CJC with DAC twice per week alone or take the CJC with DAC for a while then switch to the Ipamorelin for a while?

Subsequent studies have demonstrated the ability of different GHRPs to ameliorate local and systemic inflammatory processes in a variety of experimental scenarios by suppressing the activation of NF-κB, the consequent expression of proinflammatory cytokines, and acting as chemokine receptor antagonist [25–27]. Differentiation to myofibroblasts, collagen fibrillogenesis, and matrix accumulation are controlled by opposing forces: proinflammatory and profibrogenic, that require a fine tuning to ensure a proper esthetic healing and effective mechanical properties of the ECM [28, 29]. However, both the original GRF (1-29) and the Mod GRF 1-29 required frequent dosages. So a new compound called CJC-1295 was created which was far more stable. This compound was made by adding Lysine – which is a non-peptide, and is also called Drug Affinity Complex or DAC. Since the original Mod GRF 1-29 does not contain DAC, it is named as CJC-1295 without DAC. However, the actual CJC-1295 is not only difficult, but also very expensive to produce. GHRH (1 μg/kg) plus GHRP-6 (1 μg/kg) is given intravenously at 0 minutes and blood drawn for GH sampling at 0 and 120 minutes [200]. Ipamorelin is very similar to the growth hormone releasing peptides (GHRPs) GHRP 2 and GHRP 6 in that it mimics ghrelin (the hunger hormone) and targets a specific HGH pulse. However, unlike other GHRPs, this peptide doesn’t affect the release of cortisol, acetylcholine, prolactin and aldosterone thereby minimizing side effects experienced with other GH therapies, such as increased hunger. I have questions about combined therapy of CJC 1295 and Ipamorelin at the same time on a daily basis for both. The compounding pharmacies do not clearly state whether the CJC is with or without DAC. If it is the CJC with DAC, which sustains elevated GH and IGF-1 for several days, would taking it nightly in conjunction with the Ipramorelin, that is suggested to be taken TID but is being recommended only once at night, be over-stimulatory? If the CJC is without DAC, why take two pepetides simultaneously ,that have similar effects? I am just not clear why taking a daily dose of CJC with Ipamorelin as a single dose is better than taking the CJC with DAC twice per week alone or take the CJC with DAC for a while then switch to the Ipamorelin for a while?
GHRH/GHRP-6 was the first of a family of synthetic peptides that enhance the release of the GH by the pituitary gland in a dose-dependent manner. Since its discovery, it has been used as a benchmark and starting point for many of the research aims to obtain new drugs, but none of its implications are more engaging than the treating of the obesity epidemic.

One common concern when it comes to GHRP-6 doses (or the doses of any Ghrelin mimetic/GHRP) is the fact that it has been found to exhibit the ability to induce secretion of Cortisol and Prolactin. While many studies have indeed demonstrated this[5], they have also demonstrated that the Prolactin and Cortisol increases in most test subjects were not altered at all at GHRP-6 doses of 100mcg or less[6] [7]. GHRP-6 doses that are increased above 100mcg will exhibit increased Cortisol and Prolactin secretion, but minimally. As the dose is further increased, it stands to reason that the Cortisol and Prolactin secretions will increase as well.

GHRP-6 while being the penultimate in strengths of GH release in its class, it is still quite potent and can be taken 2-3 times in a day. It is available in a freeze-dried powder and should be reconstituted in bacteriostatic water and stored in the refrigerator. It is available in 5 mg packets, and one dosage should not be more than 100 micrograms. A dosage of more than 200 micrograms does not any significant impact on the muscles. It should be injected using an insulin syringe either under the skin or between muscles.
When taking Ipamorelin, you want it to be pushed through your system naturally, and at the same levels. If you are constantly altering the times you take it, or increase/decrease dosages during your cycle, this is not going to be attainable. To maximize the benefits and gains you are going to experience, dosage levels should be consistent, as should the timing of the dosage you are taking each day.
More than a decade ago, CD36 was identified as one of the GHRP-6 receptors [12]. This is a scavenger receptor endowed with multiligand and multifunctional capabilities and is expressed by a broad constellation of mammalian cells [13]. Granulation tissue neovascularization is perhaps the most renowned physiological role of CD36 in wound healing [14]. Serendipitous observations of our laboratory indicated that CD36 mRNA transcript appeared abundantly represented in clinical samples of granulation tissue of either acute (deep burn injuries) or chronic (pressure ulcers) wounds, as in laboratory rat’s controlled full-thickness wounds. This finding incited us to speculate on the effects associated with CD36 agonistic stimulation beyond that of the angiostatic action via thrombospondin binding [15]. Here we provide the first experimental evidence on the favorable impact of the topical administration of GHRP-6, as a candidate to qualitatively improve the healing process.
Because the ligands of most GPCRs are unknown, assays for their activity generally have no positive controls. GHS-R, however, was known to bind several artificial ligands, such as GHRP-6 or hexarelin, providing a convenient positive control for constructing the assay system used to search for the endogenous ligand. A cultured cell line expressing the GHS-R was established and used to identify tissue extracts that could stimulate the GHS-R, as monitored by increases in intracellular Ca2+ levels. After screening several tissues, very strong activity by an endogenous ligand was unexpectedly found in stomach extracts (). The ligand was finally purified by reversed-phase HPLC (RP-HPLC) and named as ghrelin. The name “ghrelin” is based on “ghre,” a word root in Proto-Indo-European languages for “grow,” in reference to its ability to stimulate GH release. Ghrelin is a 28 amino acid peptide in which the serine 3 (Ser3) is n-octanoylated and this modification is essential for the activity of ghrelin (Fig. 2). Ghrelin is the first known case of a peptide hormone modified by a fatty acid. Rat and human ghrelins differ in only two amino acid residues. There is no structural homology between ghrelin and peptide GHSs such as GHRP-6 or hexarelin.

Healthy male Wistar rats (250–270 g) were purchased from the National Center for Animal Breeding (CENPALAB, Havana, Cuba). Animals were individually housed at the animals’ facility of the Center for Genetic Engineering and Biotechnology, Havana, Cuba, and maintained under controlled environmental conditions and light cycles (12/12 hrs). Rats were fed with standard laboratory rodent’s chow under no restriction. Following an acclimation week, the dorsum of the rats was conditioned to receive two controlled full-thickness wounds, under sodium pentobarbital (30 mg/kg) anesthesia. The cuts were generated with disposable 6 mm diameter punch biotomes (Acuderm, Ft. Lauderdale, USA). Two independent experiments were performed using the above described wound model. Thus, 10 rats ( wounds) were used for either GHRP-6 formulation or vehicle (1% CMC) groups in each experiment. Upon wounds induction the rats were randomly assigned to either group. The wounds were cleansed daily with saline, their contours traced on transparent plastic sheets and treated accordingly. Treatments were topically applied twice a day at the same hours during four days. Wounds closure dynamic was measured by planimetric analysis as described previously [16] using the ImageJ software, version 1.46r. Since the GHRP-6 intervention increased the rate of closure, the animals were terminated by anesthesia overdose on day five after wounding. Ulcers and a surrounding margin of intact skin (~5 mm) were collected and hemisectioned. One hemisection was preserved in RNA Later solution for further gene expression studies. The other hemisection was fixed in 10% buffered formalin, paraffin embedded, and 5-μm sectioned. The specimens were stained with hematoxylin/eosin (H/E) and Mallory trichrome to examine collagen deposit. Other slides were destined for immunohistochemistry (as described below).

Combined, the loss of muscle and bone mass, is a quick ticket to the grave. The lack of supporting muscle and bone tissue, means that falls are more likely to occur, lengthy hospital stays inevitable, and the immobility created from these sustained injuries, produce further reduction in muscle mass and bone mass. The consumption of all dairy products have been shown to naturally raise IGF-1 levels , but I personally go straight to the source and both drink camel milk and other forms of raw milk (in moderation) and use goat’s milk colostrum.
