Growth Hormone Releasing Peptides (GHRP) are a class of compounds, which stimulate the release of growth hormone. GHRP variants include GHRP-2, GHRP-6, hexarelin, ipamorelin (Thomas et al, 2011) and agents with similar actions including CJC-1295 (Teichman et al, 2006, Acherman et al, 1999, Walker et al, 2006). These agents are considered peptide hormones. GHRPs are thought to act by stimulating the release of endogenous human growth hormone leading to pharmacological effects such as increased bone mineral density, increased lean muscle mass, modest improvements in strength and improved recovery from injuries such as fractures (Smith, 2005).
Following the preliminary histological data, suggesting a reduction of wound inflammation and a far more organized ECM, we addressed the gene expression study toward inflammatory and profibrogenic markers. We primarily examined Cd36 expression following topical GHRP-6 application and found that peptide reduced its receptor expression () (Figure 4). Furthermore, the treatment significantly reduced Adam17 expression () and approached to significantly reduce Tnf (), which may partially contribute to explaining the substantial reduction of infiltrated inflammatory cells within the wound bed (Figure 4).

Healthy male Wistar rats (250–270 g) were purchased from the National Center for Animal Breeding (CENPALAB, Havana, Cuba). Animals were individually housed at the animals’ facility of the Center for Genetic Engineering and Biotechnology, Havana, Cuba, and maintained under controlled environmental conditions and light cycles (12/12 hrs). Rats were fed with standard laboratory rodent’s chow under no restriction. Following an acclimation week, the dorsum of the rats was conditioned to receive two controlled full-thickness wounds, under sodium pentobarbital (30 mg/kg) anesthesia. The cuts were generated with disposable 6 mm diameter punch biotomes (Acuderm, Ft. Lauderdale, USA). Two independent experiments were performed using the above described wound model. Thus, 10 rats ( wounds) were used for either GHRP-6 formulation or vehicle (1% CMC) groups in each experiment. Upon wounds induction the rats were randomly assigned to either group. The wounds were cleansed daily with saline, their contours traced on transparent plastic sheets and treated accordingly. Treatments were topically applied twice a day at the same hours during four days. Wounds closure dynamic was measured by planimetric analysis as described previously [16] using the ImageJ software, version 1.46r. Since the GHRP-6 intervention increased the rate of closure, the animals were terminated by anesthesia overdose on day five after wounding. Ulcers and a surrounding margin of intact skin (~5 mm) were collected and hemisectioned. One hemisection was preserved in RNA Later solution for further gene expression studies. The other hemisection was fixed in 10% buffered formalin, paraffin embedded, and 5-μm sectioned. The specimens were stained with hematoxylin/eosin (H/E) and Mallory trichrome to examine collagen deposit. Other slides were destined for immunohistochemistry (as described below).
The scheduling of paracetamol and caffeine when combined in a compound analgesic as the only active ingredients was again reviewed by the NDPSC at its 57th Meeting in October 2009 after the Committee had received a request to reconsider the scheduling on the grounds of potential toxicity if used in excess. I weigh 215 and i have no idea what my bodyfat is, but its pretty high and not where i want it. Im currently in the middle of a strength phase, training 4 days a week Westside style. Im in the middle of week three of that, so im gonna finish my 8 week phase, training that way and see what these peps do. A peptide is an amino acid chain (amino acids being the building blocks of proteins), responsible for signalling different responses in the body. These amino chains already exist in the body in one form or another, which is why some consider them as ‘natural’ compounds (although testosterone exists in the human body too, adding extra is considered cheating in most cases). Peptides have been classified in research and manufacture according to a number sequence in many cases, which is why some are just numbers and letters (see later).
In this one you have a peptide with potential to stimulate GH release without resulting in issues associated with others. Just like GHRP-6, it both stimulates the pituitary and suppresses somatostatin. This is not the most powerful growth hormone releasing peptide. I have been using sermorelin (bioidentical growth hormone releasing hormone) for 2 months now to help heal a nasty right quad tendon rupture suffered the end of December. I’m 52 years old with 7% bodyfat and am a lifetime strength trainer and former high level bike racer. 2 months ago, in spite of months of religious rehab, I couldn’t do a single right leg bench stepup. Yesterday I was doing 20lb DB’s for repeated sets of 15. I get complete blood panels every 6 months, and my last labs in May showed my IGF-1 levels off the reference range low. The conclusion comes down to which peptides should you use. This is going to depend on personal use and experience. Whether you are into bodybuilding, gaining an edge in sports or just looking for health and wellness and anti aging properties, peptides offer a little something for everyone. There are many peptides that are not covered below, but as an introduction into the peptide world these are the most generic and widely used. As with any supplement or drug, please do your own research before diving into using peptides.
Ghrelin is a potent stimulator of growth hormone secretion from the anterior pituitary gland. The ghrelin receptor is a G protein-coupled receptor, known as the growth hormone secretagogue receptor. Ghrelin binds to the GHSR1a splice-variant of this receptor which is present in high density in the hypothalamus, pituitary as well as vagal afferent cell bodies and vagal afferent endings throughout the gastro-intestinal tract.
But gene-therapy experiments have also resulted in patient deaths. The use of such therapies can cause the human body to experience fatal immune reactions to the vectors used to place the gene in the body. Another danger of gene therapy is an inability to control the expression of the gene, which could translate into a rapidly spreading cancer. Or the expression of the gene could spread from skeletal muscle into heart muscle, resulting in excessive heart muscle growth (known as left ventricular hypertrophy, or “athlete’s heart) that can cause premature heart failure.
The response to GHSs is not gender related, except during puberty, when girls exhibit a greater response than do boys. The GH responses to both GHSs and ghrelin are similar during the early-follicular, late-follicular, and luteal phases of the menstrual cycle, suggesting that they are not affected by changes in estrogen levels. However, estrogen as well as estrogen-progestin supplementation enhances the GH response to ghrelin after menopause.
Paracetamol has long been considered very safe, without the risks of gastric injury associated with aspirin and NSAIDs. But there are distinct risks of liver injury, usually following overdose situations. In response many international regulatory authorities have taken steps to reduce the pack sizes of paracetamol, and to restrict release in some environments to pharmacies. In the USA, FDA has required prescription acetaminophen, when it is usually combined with an opioid, to reduce the dose per dose unit to 325 mg, but without reducing the maximal daily dose. No change of dosing in the USA has yet come for OTC acetaminophen. Use of paracetamol should be kept to a minimum in patients with underlying liver and renal disease. It can reduce the effects of lithium, ACE inhibitors, beta blockers and methotrexate. However, it remains one of the safest and most effective analgesic drugs, particularly in the elderly where the risks of gastric bleeding with NSAIDs are more common, and carries minimal side effects.
During studies of the opioidal control of GH secretion several analogs of met-enkephalin were found to be potent GH secretagogs. Among them were GH-releasing peptide-6 (GHRP-6), and hexarelin (His-D2MeTRP-Ala-Trp-DPhe-Lys-NH2) (Laron, 1995). They act via a receptor unrelated to that of GHRH (Howard et al., 1996). The potent biologic action of the GHRPs and the identification of a specific receptor suggested the existence of a natural ligand.
There were concerns regarding the number of contraindications and precautions and whether consumers would be able to interpret these appropriately without a requirement for pharmacist advice. There were concerns regarding gastro-intestinal, renal and other adverse effects related to the potential interactions of ibuprofen and paracetamol. Also raised were concerns regarding the potential for paracetamol overdose.

Thymosin beta 4 (Tβ4) is the predominant form of thymosin in our bodies. It has been found in high concentrations in wound tissue and certain blood cells involved in clotting, signifying its important role in the healing process. In fact, recent studies have revealed that the first gene to be upregulated after an injury is the Tβ4 gene. As the body begins the recovery process, Tβ4 aids in the creation of new vessels in the injured area, which carry blood, nutrients, and reparative substances to the site. Tβ4 also has anti-inflammatory properties, and works to decrease the amount of inflammatory substances, called cytokines. Inflammation plays a large role in many of the symptoms associated with a large number of conditions (i.e., Lyme disease, CFS, FM, autoimmune diseases, infections, etc.), making the potential impact of Tβ4 quite extensive.

IGF-1 is the only natural hormone that can stimulate lean muscle mass gains and help the body choose to burn stored fat over simple glucose for fuel, meaning, you will burn off more fat. Studies demonstrate that only colostrum supplements containing lactoferrin can produce lean muscle gains that complement IGF-1 supplementation. That’s because it is actually the lactoferrin in some brands of colostrum that work to increase muscle mass and to burn adipose tissue. In fact, in a recent 2013 study, participants who supplemented with lactoferrin over a period of eight weeks experienced increased weight loss, reduced visceral and subcutaneous fat, reduced waist circumference, and reduced hip circumference.


Results in Fig. 1.8 of normal young men (left panel) and women (right panel) demonstrate that iv bolus combined GHRP-2 and GHRH at the respective doses of 1 μg/kg GHRH and a subthreshold GH-releasing dose of 0.03 μg/kg GHRP-2 released GH synergistically (Bowers, 1998). From these studies, GHRP is envisioned to act on the hypothalamus to release an unknown factor (U factor) rather than endogenous GHRH which subsequently acts concomitantly with GHRH on the pituitary somatotroph to release GH synergistically. In this study, the important specific finding is that GHRP-2 augments GHRH release even when GHRH is present in excess amounts, and the concomitant GHRP-2 dose of 0.03 μg/kg is a subthreshold GH-releasing amount. Thus, GHRP + GHRH is not releasing GH in this study by augmenting endogenous GHRH release and, furthermore, GHRP+GHRH release in vitro is additive and not synergistic. In addition, from other high-dosage GHRP-2 data, that is, 10 μg/kg sc (not shown), we have postulated that at high doses GHRPs do act on the hypothalamus to release endogenous GHRH because high-dose GHRP-2 (10 μg/kg sc) releases the same large amount of GH released by combined GHRH + GHRP-2 at 1 + 1 μg/kg iv (Bowers, 1998a,b).
Perhaps, the primary reason why peptides are used in bodybuilding has to do with how they improve growth hormone production. It is no longer a secret how HGH helps to improve lean muscle mass. GH secretagogues differ from exogenous rHGH in their effects primarily because endogenous GH contains all five isoforms of growth hormone, whereas exogenous GH contains only the 20 kilodalton isoform. Different isoforms affect tissues in discreet ways that the 20 kDa isoform cannot. Administration of GH secretagogues causes a pulse-release of GH from the pituitary which is cleared from the body within a few hours. This does not significantly raise plasma insulin-like growth factor 1 (IGF-1) levels.[citation needed]
