GHRPs can be administered alone or in combination with GHRH. Combined administration of GHRP-6 and GHRH is the most potent stimulus to GH release, with excellent reproducibility and no serious side effects [23]. GHRH/GHRP-6 is highly specific, but is less sensitive than ITT. It is a viable alternative to the ITT in patients with organic pituitary disease, but overlap has been reported between GH levels attained in the control group and severely GH-deficient patients. For increasing GH levels, GHRP-6 is less effective in the presence of high blood glucose levels or high somatostatin levels, which result from high IGF-1 levels. For this reason, for best effect GHRP-6 should be taken while blood sugar is relatively low, for example about 30-60 minutes before a meal. GHRP-6 will have reduced effect if GH is being taken by injection, because GH increases IGF-1. Where GH use is limited to no more than about 14 IU per week, simultaneous GHRP-6 use probably still will increase GH somewhat further, but if GH use is greater than this then likely GHRP-6 injections will do little to nothing towards increasing GH levels any further.

GH-releasing peptides (GHRPs) are synthetic peptides that like GHRH act directly on pituitary somatotrophs to stimulate GH release. Growth hormone (GH) release is stimulated by a variety of synthetic secretagogues, of which growth hormone-releasing hexapeptide (GHRP-6) has been most thoroughly studied; it is thought to have actions at both pituitary and hypothalamic site.

Importantly, in the hypertrophic scars rabbit’s model, GHRP-6 intervention dramatically reduced the onset of exuberant scars by activating PPARγ and reducing the expression of fibrogenic cytokines. GHRP-6 showed no effect on the reversion of consolidated lesions. This evidence supports the notion that CD36 is an active and pharmacologically approachable receptor to attenuate wound inflammation and accelerate its closure so as to improve wound esthetic.

Anti-inflammatory: blunts NFκB expression and activation. Cell survival: it involves the phosphatidylinositol 3-kinase/RAC-alpha serine/threonine-protein kinase (PI-3K/AKT1) pathway, as the induction of the hypoxia-inducible factor-1 alpha (HIF-1α). Cardioprotective: as shown, it involves different biological actions that converge to enhance cardiomyocytes survival. Vasodilatory: it seems to involve e-NOS upregulation and endothelin activity reduction. Anabolic: it is mediated by the IGF-1/AKT1 and mTOR pathway activity.
The prescription form of IGF-1 most often injected is “mecasermin”, which goes by the trade name Increlex. Manufactured using recombinant DNA technology, mecasermin is clinically used to treat IGF-1 deficiency and stunted growth. In 1995, a potent oral GHS L-163,191 (MK-0677) was reported by Patchett et al. This agent was found to have excellent oral bioavailability and specificity in its release of GH, without significant effect on plasma levels of other hormones such as aldosterone, luteinizing hormone, thyroxine, and prolactin.
The response of these wounds reminds us of the pattern of healing described for MG53 protein (a membrane repair machinery member), so that the treatment facilitated wound healing along with a reduced scarring in rodent models. This antiscar effect was explained by interfering with TGF-β-dependent activation of myofibroblasts differentiation and reduction of ECM proteins accumulation [22]. Similarly, antiscarring healing properties are described for plants’ principles that downregulate the expression of fibrogenic-related molecules such as TGF-β1 and the downstream events, leading to fibrosis and scar formation [23]. In addition to a direct action of GHRP-6 on TGFB1 gene expression, we deem that the reduction of inflammatory effectors could have also contributed to enhancing the healing process and to reducing fibrosis. In an animal model of liver ischemia/reperfusion, we previously demonstrated that GHRP-6 prevented internal organs parenchymal activation and the onset of a systemic inflammatory response syndrome by downregulating proinflammatory cytokines [24]. Subsequent studies have demonstrated the ability of different GHRPs to ameliorate local and systemic inflammatory processes in a variety of experimental scenarios by suppressing the activation of NF-κB, the consequent expression of proinflammatory cytokines, and acting as chemokine receptor antagonist [25–27]. Differentiation to myofibroblasts, collagen fibrillogenesis, and matrix accumulation are controlled by opposing forces: proinflammatory and profibrogenic, that require a fine tuning to ensure a proper esthetic healing and effective mechanical properties of the ECM [28, 29]. The overall interpretation of the data from (i) the rate of closure, (ii) microscopic appearance of the collagen fibrils alignment/organization, (iii) impact of the treatment on the transcriptional expression of cytoskeleton filamentous proteins (smooth muscle α-actin (α-SMA), desmin, and vimentin) supports the hypothesis that, in this context, GHRP-6 has shifted the balance toward “a more regenerative” rather than a reparative phenotype.

Now, you may have heard many bodybuilders saying that when you take GHRP-6 that they get a huge and very intense increase in appetite, about 20 mins after the initial injection. Well, this is caused by the GHRP-6 antagonising the peptide Ghrelin, it mimics it, but, in reality, it actually fights against it causing the signal for gastric emptying and hunger. Ghrelin is what many believes causes obesity, and insulin resistance amongst other things, and I believe this is one way by which GHRP-6 may help reduce fat, by fighting against it. However, there is always a but, if you take more than 150mcg the effects of the gastric emptying can be so strong that you may have the urge to severely stuff yourself with food, so if you're on a bulking cycle this is a great side effect, and, considering the price, it's a very cost-effective one. Therefore, during a bulking run, I rate this as the number one aid in increasing appetite, as you also get very a good anabolic effect and increased strength.

In November 1999, the National Drugs and Poisons Schedule Committee (NDPSC) decided to reschedule mometasone from Schedule 4 to Schedule 3 for use in aqueous nasal sprays for the treatment of seasonal allergic rhinitis, with certain dose and age conditions. The NDPSC considered that this rescheduling was appropriate given mometasone's safety in use based on pharmacokinetic parameters, and that the treatment of seasonal allergic rhinitis has a place in Schedule 3.

I'm new to the forum and there is some great information here that I need to do more reading on. I've been taking Ipam and ModGrf 1-29 for at least 2yrs now. The first thing I noticed is that I have good quality sleep. I have difficulty sleeping and staying asleep. I take these peps 20min before I go to bed and get a good deep sleep for at least most of the night. I take them before doing my morning cardio, after my workout (afternoon) and before I go to bed. I've read if you take it before your morning cardio it releases more FFA to burn during your session. After the workout to aid in recovery. Before bed to aid in a deeper sleep. It is important when you take them. Dont eat before morning cardio...no carbs/sugary drinks...3hrs should have passed before taking another shot and do not eat before 15-20min after taking the peps. The reason (I've read) to wait these times is to take them while your insulin levels are low. High Insulin levels will minimize the pulsation of the GH. The phrase I've read is to try to keep "insulin quiet" to maximize the pulse of GH. Ipam can work by itself but if taken with ModGrf 1-29 it will magnify the GH Pulse. ModGrf is useless by itself. I've read that there is a saturation dose, so more is not better. 200mcg for each should do the job. Since synthetic GH shuts down natural production, these peps stimulate the pituitary to pulse more natural GH. To get the maximum effect of syn GH, you would also take the shot at the same time discussed above. Once again, this is what I've read and the protocol that I've followed. The results arent like AAS (nothing is!) but it is a good way to feel better since I cant take TRT. (trying to have kids) I also travel with the peps bc I value the sleep that I get from it.

Prolactin, or estrogen gynecomastia issues: Those that are sensitive to prolactin based gynecomastia are open to problems with GHRP-6. Luckily, this is a rare occurrence, and it can be easily combated with dostinex. However, those who stack this compound with anabolic androgenic steroids may have a greater chance of getting gynecomastia. Thats why an AI (aromatase inhibitor) should be used during this kind of cycles.
